Clinical management of skin-containing vascularized composite allografts (VCA) requires accurate assessment the graft status, typically based on skin biopsies. The Banff 2007 Working Classification proposed 4 grades acute rejection, but did not score individual features or include vascular rejection. Here we report a systematic scoring system developed from MHC-mismatched porcine VCA. Biopsies 20 VCA, 9 autologous flaps and normal were analyzed to optimize methodology set thresholds....

Dorsal spanning plate fixation can be used to treat comminuted distal radius fractures not amenable volar plating. However, there is a relative paucity of outcomes data; so, the aim this retrospective study was investigate and complications after dorsal for fractures.Distal treated with at level-1 trauma center were retrospectively identified via Current Procedural Terminology coding chart review from 2014 2019. Patient demographics, fracture pattern characteristics, techniques, clinical all...

Vascularized composite allografts (VCAs) can restore fully functional anatomic units in patients with limb amputations or severe facial tissue loss. However, acute rejection of the skin is frequently observed and underscores importance developing tolerance induction protocols. In this study, we have characterized immune system VCAs. We demonstrate infiltration recipient leukocytes, regardless status, tolerant mixed hematopoietic chimeras, co-existence these cells donor leukocytes absence...

A 31-year-old man with a history of giant cell tumor bone (GCTB) in the distal radius presents to clinic 9 years after en bloc resection. He was found have new soft tissue mass consistent GCTB and pulmonary metastases. Ultimately, he underwent excision his recurrence partial lobectomy for lung metastases. This case highlights importance having high level suspicion local or metastasis, even wide resection negative margins.

INTRODUCTION: The incidence of acute rejection (AR) the skin within first year after hand or face transplantation is approximately 85% and up to 56% patients experience multiple episodes1. Significant immunosuppression required prevent allograft loss, recent studies suggest that repeated AR episodes can lead VCA dysfunction loss2. mechanisms underlying variability in presentation remain poorly defined however. MATERIALS AND METHODS: 8 cynomolgus monkeys received either an orthotopic (n=2)...

INTRODUCTION: Tolerance in vascularized composite allotransplantation (VCA) remains elusive and patients are faced with a lifetime of immunosuppression associated risks. Tocilizumab (anti-IL-6 receptor monoclonal antibody) is currently FDA approved for use rheumatoid idiopathic arthritis. It mitigates inflammation, reduces the incidence GvHD, potentially pro-tolerogenic. We investigated utility short course tocilizumab non-human primate model (NHP) facial VCA to achieve prolonged survival...

Introduction: Microglia are resident immune cells of the central nervous system (CNS) and work to maintain a homeostatic environment allow for proper neuronal functioning. Previous studies suggest microglia originate both from neural progenitor cell population possibly myeloid progenitor. This study aims establish working model quantitatively qualitatively determine contribution hematopoietically-derived precursors CNS in non-disease state. Methods: Normal C57BL/6 mice were treated with...
