To address the limited tumor penetration of nanoparticle drug delivery vehicles, we report first pH-responsive polypeptide micelle that dissociates at low extracellular pH solid tumors. This histidine-rich elastin-like block copolymer self-assembles 37 °C into spherical micelles are stabilized by Zn2+ and disrupted as drops from 7.4 to 6.4. These pH-sensitive demonstrate better in vivo distribution tumors than a pH-insensitive control.
