Previously, we have demonstrated that the transplantation of viable, structurally intact, respiration competent mitochondria into ischemic myocardium during early reperfusion significantly enhanced cardioprotection by decreasing myocellular damage and enhancing functional recovery. Our in vitro vivo studies established autologous are internalized cardiomyocytes following transplantation; however, mechanism(s) modulating internalization these organelles were unknown. Here, show occurs through...
Previously described mitochondrial isolation methods using differential centrifugation and/or Ficoll gradient require 60 to 100 min complete. We describe a method for the rapid of mitochondria from mammalian biopsies commercial tissue dissociator and filtration. In this protocol, manual homogenization is replaced with dissociator's standardized cycle. This allows uniform consistent that not easily achieved homogenization. Following dissociation, homogenate filtered through nylon mesh...
The role of mtDNA and nuclear genome in human aging was examined by their intercellular transfer using skin fibroblasts mtDNA-less HeLa cells (rho o-HeLa cells). We found vivo age-related reductions the activity cytochrome c oxidase obtained from 16 donors various ages (0-97 years). abnormality mitochondria aged not attributable to either decrease copy number molecules or increase deletion mutant molecules, but significant overall polypeptide synthesis mitochondria. However, experiments...
Adult liver malignancies, including intrahepatic cholangiocarcinoma and hepatocellular carcinoma, are the second leading cause of cancer-related deaths worldwide. Most individuals treated with either combination chemotherapy or immunotherapy, respectively, without specific biomarkers for selection. Here using high-throughput screens, proteomics in vitro resistance models, we identify small molecule YC-1 as selectively active against a defined subset cell lines derived from both cancer types....
When the alkB gene of Escherichia coli that controls sensitivity bacteria to methyl methanesulfonate was placed under control lac regulatory region on a multicopy plasmid, product, AlkB protein, overproduced. By monitoring band sodium dodecyl sulfate-polyacrylamide gel electrophoresis, protein purified near physical homogeneity. An amino-terminal sequence and total amino acid composition were in accord with deduced from nucleotide gene, determined by phage M13 dideoxy method. It concluded is...
We addressed the question of whether both mitochondrial and cytoplasmic translation activities decreased simultaneously in human skin fibroblasts with age donors found that age-related reduction was limited to translation. Then, determine which genome, or nuclear, responsible for this age-related, mitochondria-specific reduction, pure nuclear transfer carried out from DNA (mtDNA)-less HeLa cells four fibroblast lines, two aged subjects, one a fetus, patient cardiomyopathy, their hybrid...
Effects of ketamine on the sodium (INa) and L-type calcium currents (ICa) were examined by using whole-cell patch clamp techniques in guinea pig single ventricular myocytes. The mode action was compared with those quinidine, a channel blocker, verapamil, blocker. Ketamine (30-300 μM) inhibited both INa ICa concentration-dependent manner. Quinidine (30 verapamil (0.1 produced use-dependent depression ICa, respectively. amplitude elicited first depolarizing pulse after long quiescent period...

To evaluate the significance of expression vascular endothelial growth factor (VEGF), its correlation with clinicopathological variables were studied in tissue hepatocellular carcinoma (HCC) and surrounding liver. In 56 samples (tumor non-tumor liver tissue) collected from 28 patients, VEGF was examined by immunohistochemistry western blot analysis. The value blotting correlated immunohistochemical staining grade. In tumor tissue, size (P = 0.034), á-fetoprotein 0.036) protein induced vitamin...
The inotropic effect of diazepam, a benzodiazepine derivative, and its mechanism action were examined using guinea pig heart single ventricular cell preparations. In Langendorff hearts right free-wall preparations, diazepam (10 to 100 microM) produced monophasic negative in concentration dependent manner. Neither central type (flumazenil 1 nor peripheral (PK11195 10 receptor antagonist antagonized the effects diazepam. Diazepam shortened potential duration papillary muscle isolated cells,...
Gadolinium-enhanced dynamic magnetic resonance (MR) images in 90 patients were reviewed to assess the artifacts mimicking portal venous thrombosis. The incidence of definite signal-intensity decrease pathologic condition was higher (P < 0.01) right (8%) and left (9%) vein branches trunk (6%) than splenic (0%) or superior mesenteric (1%) with equilibrium-phase images. Radiologists should remember that MR occasionally show thrombosis due flow artifact.
