cDNA clones encoding three classes of human actins have been isolated and characterized. The first two (gamma beta, cytoplasmic actins) were obtained from a library constructed simian virus 40-transformed fibroblast mRNA, the third class (alpha, muscle actin) was adult mRNA. A new approach developed to enrich for full-length cDNAs. plasmid linearized with restriction enzymes that did not cut inserts interest; it then size-fractionated on gels, chimeric molecules optimal length selected...

We report the complete nucleotide sequence of a human β actin cDNA. Both 5′ and 3′ untranslated regions are similar (>80%) to analogous rat β-actin gene reported by Nudel et al (1983) 1 . When segment region is used as radiolabelled probe, strong hybridization chick mRNA seen. This conservation sequences suggests that selective pressures operate on non-translated segments mRNA.

We have constructed isotype-specific subclones from the 3' untranslated regions of alpha-skeletal, alpha-cardiac, beta-cytoskeletal, and gamma-cytoskeletal actin cDNAs. These clones been used as hybridization probes to assay number organization these isotypes in human genome. Hybridization genomic (Engel et al., Proc. Natl. Acad. Sci. U.S.A. 78:4674-4678, 1981; Engel Mol. Cell. Biol. 2:674-684, 1982) has allowed unambiguous assignment isotypically defined subfamilies. In addition, only one...

We have assigned six members of the human beta-actin multigene family to specific chromosomes. The functional gene, ACTB, is located on chromosome 7, and other beta-actin-related sequences are dispersed over at least four different chromosomes including one locus X chromosome. A common nonsense polymorphism (R577X) in the ACTN3 gene results complete deficiency of fast skeletal muscle fiber protein α-actinin-3 an estimated one billion humans worldwide. The XX null genotype is under-represented elite sprint athletes, associated with reduced strength and performance non-athletes, over-represented endurance suggesting that increases at cost power generation. Apoptosis is an important biological process required for the removal of unwanted or damaged cells. Mounting evidence implicates actin cytoskeleton as both a sensor and mediator apoptosis. Studies also suggest that binding proteins (ABPs) significantly contribute to apoptosis dynamics play key role in regulating signaling. The specific functions of greater than 40 vertebrate nonmuscle tropomyosins (Tms) are poorly understood. In this article we have tested the ability two Tm isoforms, TmBr3 and human homologue Tm5 (hTM5 NM1 ), to regulate actin filament function. We found that these Tms can differentially alter organization, cell size, shape. hTm5 was able recruit myosin II into stress fibers, which resulted in decreased lamellipodia cellular migration. contrast, transfection induced lamellipodial formation,...
