The article presents information regarding the metabolism of carbohydrates and minerals in Romanov sheep Kemerovo region. correlations among macro elements carbohydrate indices heavy metal levels organs tissues were ascertained. We determined average content, variability reference intervals glucose, amylase, lactate dehydrogenase blood serum. investigated relationship between these tissues. In breeding zone, content was analyzed soil, water, feed did not exceed MAC. determination macroand...

Ion pump, Na,K‐ATPase specifically binds cardiotonic steroids (CTS), which leads to inhibition of the enzyme activity and activation signaling network in cell. We have studied interaction with CTS two different types – marinobufagenin ouabain. shown that both inhibit same K i values, but binding ouabain is sensitive conformation while not. Furthermore, results structural changes Na,K‐ATPase. Our data allow explain diversity effects on receptor function caused by CTS.

Abstract Recent studies demonstrated that in addition to Na + ,K -ATPase inhibition cardiotonic steroids (CTSs) affect diverse intracellular signaling pathways. This study examines the relative impact of [Na ] i /[K -mediated and -independent transcriptomic changes triggered by endogenous CTSs ouabain marinobufagenin (MBG) human umbilical vein endothelial cells (HUVEC). We noted prolongation incubation increased apparent affinity for estimated loss [K gain . Six hour exposure HUVEC 100 3,000...

The structural organization of natural pigment-protein complexes provides a specific environment for the chromophore groups. Yet, proteins are inherently dynamic and conformationally mobile. In this work, we demonstrate heterogeneity chromophores C-phycocyanin (C-PC) from Arthrospira platensis. Part population trimeric C-PC is subject to spontaneous disturbances protein–protein interactions resulting in increased conformational mobility chromophores. Upon fluorescence excitation visible...

Active transport of sodium and potassium ions by Na,K-ATPase is accompanied the enzyme conformational transition between E1 E2 states. ATP ADP bind to in conformation with similar affinity but properties complexes these nucleotides are different. We have studied thermodynamics binding adenine at different temperatures using isothermal titration calorimetry. Our data indicate that β-phosphate involved complex formation increasing an order magnitude, while γ-phosphate does not affect it....

Sodium-potassium adenosine triphosphatase (Na,K-ATPase) creates a gradient of sodium and potassium ions necessary for the viability animal cells, it is extremely sensitive to intracellular redox status. Earlier we found that regulatory glutathionylation determines Na,K-ATPase sensitivity but role basal other modifications cysteine residues not clear. The purpose this study was detect oxidized, nitrosylated, or glutathionylated in Na,K-ATPase, evaluate possibility removing these assess their...

Prolonged hyperosmotic shrinkage evokes expression of osmoprotective genes via nuclear factor NFAT5-mediated pathway and activates Na+ influx hypertonicity-induced cation channels (HICC). In human umbilical vein endothelial cells (HUVEC) elevation intracellular sodium concentration ([Na+]i) triggers transcription dozens early response (ERG). This study examined the role monovalent cations in Na+i-sensitive ERGs iso- hyperosmotically shrunken HUVEC.Cell volume was measured by 3D...

Cardiotonic steroids (CTSs) are specific inhibitors of Na,K-ATPase (NKA). They induce diverse physiological effects and were investigated as potential drugs in heart diseases, hypertension, neuroinflammation, antiviral cancer therapy. Here, we compared the inhibition mode binding CTSs, such ouabain, digoxin marinobufagenin to NKA from pig rat kidneys, containing CTSs-sensitive (α1S) -resistant (α1R) α1-subunit, respectively. Marinobufagenin contrast ouabain interacted with α1S-NKA...
