A new affinity column for renin was prepared by coupling the isosteric peptide inhibitor of renin, H.77 (D-His-Pro-Phe-His-LeuR-Leu-Val-Tyr, where R is a reduced bond, -CH2-NH-), to activated 6-aminohexanoic acid-Sepharose 4B. Chromatography crude extract human kidney cortex on this material resulted in 5500-fold purification 76% yield. The purified enzyme (specific activity 871 units/mg) free non-specific acid-proteinase and stable at pH 6.8 -20 degrees C over period several weeks.
Three experiments are described using new substrate analogue inhibitors of renin. The first experiment shows that introduction a reduced isostere in the scissile peptide bond an greatly increases its ability to inhibit renin particular species. However, different species have amino acids their and variation here also influences affinity hence analogues. Finally, substitution C-terminal adjacent efficacy analogues as inhibitors. In our second inhibitor dog renin, H.77, was used column produce...
A six-month project was undertaken in the Portsmouth Healthcare NHS Trust to identify prevalence of pressure sores and assess current practice level knowledge among nurses working community. Questionnaires were returned by 99 sore found be 4.4%; 72% respondents performed risk assessment routinely, 49% had access appropriate equipment 60% good or acceptable levels knowledge. However, 40% poor factors for development, 80% did not routinely carry out mattress testing. Eighty-five per cent...
Congenital hypothyroidism is screened for in the UK using blood spot thyroid-stimulating hormone (TSH) screening at 5–8 d of age. Although standards are set by Newborn Screening Programme Centre, there variations TSH cut-offs used. The introduction repeat preterm babies 36 weeks' gestational age 2005 was controversial its utility and timing. Two cases presented, who had normal values on first test became screen positive when re-tested term. with Trisomy 21 born 29 + 6 weeks an initial 3.3...
Synthetic Peptide Hormones, Human Big Gastrin I. A new total synthesis of the tetratriacontapeptide amide corresponding to proposed primary structure human big gastrin I is described. The synthetic route was based on preparation six suitably protected fragments, related sequence 28-34, 23-27, 21-22, 15-20, 9-14, and 1-8, be used as building blocks for synthesis. protecting groups were selected according Schwyzer-Wünsch strategy maximum side chain protection tertiary alcohols, also imidazol...
Abstract Fulminant endocarditis affecting the mitral valve in an 11-year-old boy was caused by a nontoxogenic strain of Corynebacterium diphtheriae.
To aid in the design of an effective inhibitor to human renin, it is essential have a detailed knowledge how this aspartic proteinase interacts with its substrate, angiotensinogen. Human renin shows stringent specificity toward Leu–Val bond natural substrate. The minimal length for substrate has been characterised as octapeptide sequence derived from amino terminal portion angiotensinogen (residues 6 — 13): His–Pro–Phe–His–Leu–Val–Ile–His (Leu–Val scissile bond). This suggests that fairly...
Summary: Infusion of H.261, the inhibitor human renin in baboon, lowered blood angiotensin I, plasma II, and arterial pressure suggesting that sodium-depleted state II contributes to maintenance pressure. In a second experiment dose-response infusions were given conscious dogs before during infusion H.77. These suggested contribution this was made mainly by circulating peptide. Preliminary results normal humans show H.142 intravenously

Chemical modification of the backbone at cleavage site in (6-13)-octapeptide equine angiotensinogen resulted greatly increased binding affinity and resistance to by renin. The D-His6-Tyr13 octapeptide analog containing reduced bond -CH2-NH-instead a peptide -CO-NH- Leu10-Leu11 linkage (H-77) was powerful vitro inhibitor canine renin (IC50 = 24nM). It gave an IC50 1 microM against human 0.6 rat In sodium-depleted conscious dogs, infusion H-77 caused dose-related falls plasma angiotensin I II...
Abstract Background and aim: Recent published evidence suggests that digital infrared breast scanning may be a useful adjunctive tool for cancer detection. In this study we have assessed the effectiveness of imaging captured using Sentinel BreastScan (Infrared Sciences Corp., Bohemia, NY, USA) compared computer-generated reports from system with expert manual review images as well new software programme currently under development called NoTouch BreastScan. Methods: A total 100 patients...
