目的 研究细胞色素b基因(Cytb)突变与柔嫩艾美耳球虫(Eimeria tenella)对喹啉类抗球虫药产生抗药性的关系。 方法 收集安徽和县、舒城县、肥东县、庐江县、凤台县和全椒县6个地区的柔嫩艾美耳球虫, 经分离、纯化、鉴定和鸡体试验法检测耐药性后, 采用PCR法和DNA测序相结合的方法, 分析各个地区的野外分离株及敏感株之间Cytb基因的序列突变情况。 结果 获得了1 100 bp的目的片段。测序后发现:与敏感株相比, 耐药株中Cytb基因有更多的基因突变, 主要表现为T→C、T→A、A→G的突变, 其中以T→A的突变最为明显。其中一些碱基的突变引起了氨基酸的改变。 结论 这些碱基的突变有可能是导致柔嫩艾美耳球虫产生对癸氧喹酯抗药性的原因之一。
