ABSTRACT Members of the Closteroviridae and Potyviridae families plant positive-strand RNA viruses encode one or two papain-like leader proteinases. In addition to a C-terminal proteolytic domain, each these proteinases possesses nonproteolytic N-terminal domain. We compared functions several using gene swapping approach. The proteinase (L-Pro) Beet yellows virus (BYV; closterovirus) was replaced with L1 L2 Citrus tristeza (CTV; another closterovirus), P-Pro Lettuce infectious (LIYV;...

Significance Epstein–Barr Virus (EBV) genome persistence and expression in human cells can result lymphocyte or epithelial cell malignancies. EBV gene depends on the encoded virus nuclear antigen 1 (EBNA1) protein binding to cognate sites episomes. Search for a necessary EBNA1 interaction with episomes identified low-level nucleolin (NCL) association EBNA1. Despite stable association, NCL shRNA knockdowns revealed wild-type NCL, but not an ATP site point mutant be critical episomes,...

Epstein-Barr Virus (EBV) is an oncogenic γ-herpesvirus that capably establishes both latent and lytic modes of infection in host cells causes malignant diseases humans. Nuclear antigen 2 (EBNA2)-mediated transcription cellular viral genes essential for the establishment maintenance EBV latency program B lymphocytes. Here, we employed a protein affinity pull-down LC-MS/MS analysis to identify nucleophosmin (NPM1) as one proteins bound EBNA2. Additionally, specific domains are responsible...

Epstein-Barr Virus (EBV) Nuclear Antigen 1 (EBNA1)-mediated origin of plasmid replication (oriP) DNA episome maintenance is essential for EBV-mediated tumorigenesis. We have now found that EBNA1 binds to Ribosome Protein L4 (RPL4). RPL4 shRNA knockdown decreased activation an oriP luciferase reporter, binding in lymphoblastoid cell lines, and EBV genome number per line. infection increased expression redistributed nuclei. Nucleolin (NCL) were a scaffold EBNA1-induced complex. The N terminus...

Epstein-Barr virus (EBV) nuclear antigen 2 (EBNA2) plays an important role in driving immortalization of EBV-infected B cells through regulating the expression many viral and cellular genes. We report a structural study tumor suppressor BS69/ZMYND11 C-terminal region, comprised tandem coiled-coil-MYND domains (BS69CC-MYND), complex with EBNA2 peptide containing PXLXP motif. The coiled-coil domain BS69 self-associates to bring two separate MYND close proximity, thereby enhancing MYND-EBNA2...

The 66-kDa leader proteinase (L-Pro) of the Beet yellows virus (BYV) possesses a nonconserved N-terminal domain and conserved, papain-like C-terminal domain. Previous work revealed that functions in RNA amplification, whereas is required for autoproteolysis. Alanine-scanning mutagenesis was applied to complete functional analysis L-Pro throughout life cycle. This indicated L-Pro, addition being proteolysis, also amplification these two are genetically separable. Examination role BYV...

ABSTRACT Epstein-Barr virus nuclear antigen (EBNA) leader protein (EBNALP) is one of the first viral genes expressed upon B-cell infection. EBNALP essential for EBV-mediated immortalization. thought to function primarily by coactivating EBNA2-mediated transcription. Chromatin immune precipitation followed deep sequencing (ChIP-seq) studies highlight that frequently cooccupies DNA sites with host cell transcription factors (TFs), in particular, EP300, implicating a broader role regulation. The Epstein-Barr virus nuclear leader protein LP (EBNALP) and EBNA2 are expressed first in lymphocyte infection, coordinately regulate cell viral gene transcription, critical for outgrowth into lymphoblastoid lines (LCLs). The beet yellows closterovirus leader proteinase (L-Pro) possesses a C-terminal domain and nonproteolytic N-terminal domain. It was found that although L-Pro is not essential for basal-level replication, deletion of its resulted in 1, 000-fold reduction RNA accumulation. Mutagenic analysis the revealed structural flexibility except 54-codon-long, 5'-terminal element corresponding open reading frame critical efficient amplification at both protein levels.

The colors portions can be enhanced or modified by PL for assistance in reducing intrasexual and male harassment, improving mature mating conspecific identity. Therefore, that contribute color modification coloration are involved cuticle evolution damselflies. micro-PL...
