We herein describe a flexible synthesis of small library 68Ga-labeled CAIX-targeted molecules via an orthogonal 2-cyanobenzothiazole (CBT)/1,2-aminothiol click reaction. Three novel CBT-functionalized chelators (1⁻3) were successfully synthesized and labeled with the positron emitter gallium-68. Cross-ligation between pre-labeled bifunctional (BFCs) 1,2-aminothiol-acetazolamide derivatives (8 9) yielded six new CAIX ligands high radiochemical yields. The reaction conditions optimized to...

A new synthetic approach toward the bacterial transglycosylase substrates, Lipid II (1) and IV (2), is described. The key disaccharide was synthesized using concept of relative reactivity value (RRV) elaborated to by conjugation with appropriate oligopeptides pyrophosphate lipids. Interestingly, results from our HPLC-based functional TGase assay suggested has a higher affinity for enzyme than II.

The development of iminocyclitol-based small molecule libraries against a bacterial TGase is described. An iminocyclitol was conjugated with pyrophosphate mimic using either 1,3-dipolar cycloaddition or reductive amination reaction, which then condensed variety lipophilic carboxylic acids in an amide bond coupling to generate desired molecular library. With assistance microtiter plate-based combinatorial chemistry and situ screening, potential inhibitor, the first potent inhibitor TGases...

Abstract A flexible synthetic strategy toward the preparation of diverse N ‐substituted muramyl dipeptides ( MDPs) from different protected monosaccharides is described. The MDPs include ‐acetyl MDP and ‐glycolyl MDP, known NOD2 ligands, this methodology allows for structural variation at six positions, including muramic acid, peptide, moieties. capacity these molecules to activate human in innate immune response was also investigated. It found that addition methyl group C 1 position...

A feasible synthetic approach toward the Mycobacterium tuberculosis (Mtb) N-glycolyl lipid II-like molecule 1 is described. Compound bears pendant undecaprenol and l-lysin moieties instead of naturally occurring decaprenol meso-diaminopimelic acid, which are not readily available. Functionalization with a fluorophore on peptide side chain gave 14, was found to be recognized as an Mtb TGase substrate. This result suggests it has tremendous utility for mechanistic studies, characterization...

Breaking down barriers: A rapid, inexpensive preparation of the structurally complex mycobacterial N-glycolyl Lipid I, II, and their analogues from a range different synthetic N-glycinyl Park's nucleotides is described (see scheme). The biotransformations were catalyzed by readily available biocatalyst obtained bacterial cell-free membrane fraction. unnatural II was found to be substrate Mycobacterium tuberculosis (Mtb) transglycosylase, PonA, I weak inhibitor against PonA. An open tip rhinoplasty was used to correct moderate severe secondary cleft lip nasal deformities in 122 patients from January 1986 1988. The results of the surgery on 92 who were followed for 1 year or more evaluated. When performed after age 3 years, 60% had a satisfactory result. pathology deformity included 32% with deficiencies lining, 35% needed framework support such as columellar strut, and 45% required correction deficiencies. Patients using postoperative stent 71% result compared...
