A large series of assays the hepatocarcinogenic potential 112 different compounds werecarried out using a rapid bioassay system developed in this laboratory based on two-step concept hepatocarcinogenesis. Rats were initally given single dose (200 mg/kg) diethylnitrosamine (DEN) i.p. and starting 2 weeks later treated with test for 6 then killed, all rats being subjected to two-thirds partial hepatectomy (PH) at week 3.Carcinogenic was scored by comparing number area per cm induced...

Urinary bladder carcinogenesis associated with melamine treatment was examined concomitant use of NaCl to allow assessment the relationship between uroliths and lesion development Analysis chemical composition calculi also performed. F344/DuCrj male rats received diets containing 3 or 1% alone in combination either 10 5% NaCl, 10% for 36 weeks, then diet without supplement a further 4 weeks. The water intake, used as an index urinary output, increased by treatment. incidences transitional...
The dose-dependence of 2-amino-1-methyl-6-phenylimidazo-[4,5-b]pyridine (PhIP) carcinogenicity was investigated in F344 rats both sexes administered the heterocyclic amine diet at concentrations 25 or 100 p.p.m. for up to 104 weeks. Incidences mammary adenocarcinomas were 7% (2/30) and 47% (14 30 rats) females those colon 43% (13/30) males 13% (4/30) groups. No induced no carcinomas observed groups either sex. Furthermore, development lymphocytic leukemia apparently enhanced by PhIP males....
The potential of purple sweet potato color (PSPC) and red cabbage (RCC), natural anthocyanin food colors, to modify colorectal carcinogenesis was investigated in male F344/DuCrj rats, initially treated with 1,2-dimethylhydrazine (DMH) receiving 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP) the diet. After DMH initiation, PSPC RCC were given at a dietary level 5.0% combination 0.02% PhIP until week 36. No or RCC-treatment-related changes clinical signs body weight found. Incidences...

Studies were made on the dose response of saccharin in induction bladder lesions. Inbred F344 rats both sexes pretreated with 100 ppm N-butyl-N-(4-hydroxybutyl)nitrosamine (BBN) drinking water for 4 weeks. Sodium was given at 50,000, 10,000, 2,000, and 400 diet 32 weeks after BBN administration, surviving killed end week 36 experiment. No increase incidence papilloma or cancer noted either sex any as compared to levels induced by alone. The incidences two types hyperplasia average number...
Studies were made on the effects of butylated hydroxyanisole (BHA), ethoxyquin (EQ) and acetaminophen (AAP) induction neoplastic lesions in liver kidney rats initiated by N-ethyl-N-hydroxyethylnitrosamine (EHEN). The number area histochemical gamma-glutamyltranspeptidase-positive (gamma-GT+) foci per unit section given BHA, EQ or AAP significantly less than EHEN alone. Similarly, hyperplastic nodules (HN) groups BHA their control groups. Induction hepatocellular carcinoma (HCC) was also...
The effects of pre- (initiation stage) and post- (promotion administration 17 chemical carcinogens (including both hepatic non-hepatic carcinogens), 3 promoters 2 non-carcinogenic analogs, on the induction liver hyperplastic nodules were investigated in rats. Test chemicals administered during initiation stage after which rats fed dietary N-2-fluorenylacetamide (2-FAA) for weeks conjugation with necrogenic CCl 4 to enhance production initiated by test chemicals. Alternatively, promotion...
We previously found that genetic polymorphism in cytochrome P450 2A6 (CYP2A6) is one of the potential determinants tobacco-related lung cancer risk. It has been reported plasma concentration cotinine, a major metabolite nicotine, carriers wild-type alleles CYP2A6 considerably higher than null or reduced-function CYP2A6, raising possibility cotinine plays an important role development cancer. As novel mechanism tumorigenesis mediated by we investigated effects on suppression apoptosis and...
The chemopreventive efficacy of lycopene and curcumin with regard to prostate carcinogenesis was investigated using 3,2'-dimethyl-4-aminobiphenol (DMAB)- 2-amino-1-methylimidazo[4,5-b]pyridine (PhIP)-induced rat ventral cancer models. Three 60 week experiments male F344 rats were carried out. In the first DMAB given for 20 weeks or administered concomitantly subsequently at dietary doses 15 500 p.p.m., respectively. second experiment pretreated 5, 45 p.p.m. 100 third PhIP selected as an...
For a long period, it has been generally considered that carcinogens, particularly genotoxic ones, have no threshold in exerting their potential for cancer induction. However, the non‐threshold theory can be challenged with regard to assessment of risk humans. Here we show food‐derived, hepatocarcinogen, 2‐amino‐3,8‐dimethylimidazo[4,5‐ f ]quinoxaline, forms DNA adducts at low doses, but does not induce glutathione S‐transferase placental form (GST‐P)‐positive foci (considered preneoplastic...
