The complete amino acid sequence of the porcine cardiac muscarinic acetylcholine receptor has been deduced by cloning and sequencing cDNA. tissue location RNA hybridizing with cDNA suggests that this species represents M 2 subtype.
The vertebrate olfactory system discriminates a wide variety of odorants by relaying coded information from sensory neurons in the epithelium to cortical areas brain. Recent studies have shown that first step odor discrimination is mediated approximately 1000 distinct receptors, which comprise largest family G-protein-coupled receptors. In present study, we used Ca(2+) imaging and single-cell reverse transcription-PCR techniques identify mouse responding an odorant subsequently clone...
We have identified novel G protein-coupled receptors (GPCRs) with no introns in the coding region from human genome sequence: 322 olfactory receptors; 22 taste 128 registered GPCRs for endogenous ligands; 50 GPCR candidates homologous to and 59 not GPCRs. The total number of without was estimated be approximately 950, which 500 are odorant or 450 ligands.
The olfactory system is remarkable in its capacity to discriminate a wide range of odorants through series transduction events initiated receptor neurons. Each neuron expected express only single odorant gene that belongs the G protein coupled family. ligand–receptor interaction, however, has not been clearly characterized. This study demonstrates functional identification receptor(s) for specific odorant(s) from neurons by combination Ca 2+ -imaging and reverse transcription–coupled PCR...
A novel variant of the S49 mouse lymphoma has been selected from wild-type cells by growth in medium containing beta-adrenergic agonist terbutaline and inhibitors cyclic nucleotide phosphodiesterase. In contrast to situation clone, synthesis adenosine 3':5'-monophosphate (cyclic AMP) is not stimulated agonists or prostaglandin E1 either intact membrane preparations such clones. However, basal NaF-stimulated activities adenylate cyclase [ATP pyrophosphate-lyase (cyclizine), EC 4.6.1.1] are...
The GTP binding regulatory protein (Ni involved in adenylate cyclase inhibition was purified from rat brain and reconstituted, together with muscarinic cholinergic receptors porcine brain, into phospholipid vesicles. Guanosine 5'-O-(3-[35S]thio)-triphosphate ([35S]GTP gamma S) hydrolyzing activities of reconstituted Ni were stimulated by the addition a agonist, carbachol. effect carbachol to increase Vmax values these activities, but Km also increased slightly most cases. Carbachol bound...
Abstract— Synaptosomes took up [ 14 C]choline, about half or more of which was converted to I4 C]acetylcholine when incubated in an appropriate medium containing 1 5 μ M‐[ C] choline and neostigmine. The amount synthesized synaptosomes increased parallel with the increase Na + concentration incubation medium. effect on uptake C]choline into dependent About 25 per cent released rapidly by increasing K from m 35 . change hardly affected release C]acetylcholine. rather small compared that...
Sequestration of m2 receptors (muscarinic acetylcholine receptor subtypes), which was assessed as loss N-[3H]methylscopolamine ([3H]NMS) binding activity from the cell surface, examined in COS 7 and BHK-21 cells that had been transfected with expression vectors encoding and, independently, a G protein-coupled kinase (GRK2) (beta-adrenergic 1) or GRK2 dominant-negative mutant (DN-GRK2). The sequestration became apparent when were treated 10(-5) M higher concentrations carbamylcholine. In this...
Muscarinic acetylcholine receptors purified from porcine brain were reconstituted with two kinds of GTP-binding proteins (Gi and Go).The binding agonists was affected by guanine nucleotides when the receptor either Gi or Go, but not in absence one proteins. The displacement curves for [ 3 H ] q ~i n ~clidinyl benzylate ( [3H]QNB) explained assuming there are sites different affinities a given agonist. The proportion high affinity site increased increasing concentrations proteins, maximum value...
The muscarinic acetylcholine receptor of porcine cerebrum has been purified to apparent homogeneity by affinity chromatography, with conjugated 3-(2'-aminobenzhydry1oxy)tropane (ABT) as described previously (Haga, K., and Haga, T. (1983) J. Biol.Chern.258, 13575-13679).In a single step purification using 900 ml digitonin/cholate-solubilized preparations 300 the ABT-agarose gel, we obtained, in yield 10-15%, more than 250 pmol receptors which bind [3H]N-methylscopolamine specific activity...
The primary structure of the α‐subunit adenylate cyclase‐inhibiting G‐protein (g i ) has been deduced from nucleotide sequence cloned DNA complementary to bovine cerebral mRNA encoding polypeptide. A much higher degree amino acid homology is observed between α‐subunits G and transducin (68%) than those cyclase‐stimulating (G s (43%) or transucin (42%).
A novel hypernodulation mutant line was isolated from Lotus japonicus Miyakojima MG-20 by irradiation with a helium ion beam. This mutant, named klavier (klv), had roots that were densely covered small nodules. The nodulation zone of klv significantly wider than the wild type. Grafting experiments showed is impaired in long-distance shoot-to-root autoregulatory mechanism. Thus shoot genotype found to be responsible for negative regulation nodule development KLV. Nodulation higher tolerance...
1. Either l-[4,5-(3)H]leucine or [Me-(3)H]choline, both l-[U-(14)C]leucine and [Me-(3)H]-choline, were injected into the ninth dorsal root ganglion of frog, peripheral transport labelled proteins and/or phospholipids, mostly phosphatidylcholine, was studied by analysis consecutive segments sciatic nerve. 2. At 25 degrees C, approx. 5% (3)H-labelled protein transported at rate 152mm/day. The temperature-dependent with Q(10) value 2.6. flow completely blocked local application colchicine, but...
Na + ‐dependent, high‐affinity choline uptake in cholinergic neurons is the rate‐limiting step acetylcholine synthesis. Here we report molecular cloning and functional characterization of human transporter (hCHT1). The hCHT1 exhibits significant homology with known members ‐dependent glucose family, but not neurotransmitter family. CHT1 gene 25 kb length 9 exons was assigned to chromosome II at position IIq11–12. Northern blot analysis showed that a 5.4 transcript expressed exclusively...
We examined the agonist‐dependent sequestration/internalization of dopamine D2 receptor (the long form D2L and short D2S), which were transiently expressed in COS‐7 HEK 293 cells with or without G‐protein‐coupled kinases (GRK2 GRK5). Sequestration was assessed quantitatively by loss [ 3 H] sulpiride‐binding activity from cell surface transfer spiperone‐binding membrane fraction to light vesicle sucrose‐density gradients. In expressing receptors alone, virtually no sequestration observed...
