The aromatase-knockout (ArKO) mouse provides a useful model to examine the role that estrogens play in development and homeostasis mammals. Lacking functional Cyp19 gene, which encodes aromatase, ArKO cannot synthesize endogenous estrogens. We examined adipose depots of male female mice, observing these animals progressively accumulate significantly more intraabdominal tissue than their wild-type (WT) littermates, reflected increased adipocyte volume at gonadal infrarenal sites. This...

It is well established that spermatogenesis controlled by gonadotrophins and testosterone. However, a role for estrogens in male reproduction recently was suggested adult mice deficient estrogen receptor α. These became infertile primarily because of an interruption fluid reabsorption the efferent ductules epididymis, thus leading to disruption seminiferous epithelium [Hess, R. A., Bunick, D., Lee, K. ABSTRACT Organizations cannot function effectively if their employees do not follow organizational rules and policies. In this paper, we explore why how in two high‐tech organizations often broke or bent designed to protect employers' confidential information (CI). The CI protection sometimes imposed requirements that disrupted employees' work, forcing choose between rule compliance doing work efficiently. Employees these situations the them ways enabled meet some of rules' requirements,...

Research has pointed to the importance of interpersonal relationships in meaningfulness work, but significant and under-examined issues remain. For example, what types have potential produce a sense work? Further, through mechanisms do these relationship features affect To answer questions, we adopt social networks theory lens explore how content structure an individual's network ties can influence his or her experience at work. Our central argument is that various resources facilitate...

Previous studies employing the male aromatase knockout (ArKO) mouse have indicated that local expression of estrogens appears to be important for progression spermatogenesis. In absence estrogen biosynthesis round spermatids are observed undergo apoptosis and thus fail differentiate into mature, elongated spermatids. This lesion arise between ages 18 weeks 1 year. To ultimately determine if disruption spermatogenesis arises earlier than weeks, we performed an intensive study examine...

Estrogen is synthesized in the testis, both Leydig cells and seminiferous epithelium, its importance spermatogenesis highlighted by phenotype of aromatase knockout (ArKO) mouse. These mice are unable to synthesize endogenous estrogens. The males develop postmeiotic defects 18 wk age. We hypothesized that maintenance younger animals may be mediated exogenous estrogenic substances. Dietary soy meal, contained almost all commercial rodent diets, provides a source isoflavones. thus investigated...

The liver X receptor (LXR)α and -β has been found to play a central role in maintaining cellular cholesterol homeostasis. In this study we comprehensively investigated the effect of deleting LXRα on testicular morphology function. absence LXRβ, excessive accumulated Sertoli cells from 2.5 months, resulting severe disruption dysregulation spermatogenesis by 10 months age. This correlated with gene expression analyses that clearly indicated LXRβ was dominant transcript testis Although LXRα−/−...

Abstract The liver × receptor (α,β) is responsible for regulating cholesterol homeostasis in cells. However, our studies using the LXRα−/−, LXRβ−/−, and LXRα−/−β−/− mice show that both LXRα β are also important bone turnover, mainly by osteoclast differentiation/activity. Introduction: receptors primarily within cells whole body. as recent role this expanding, we studied whether LXRs could be implicated development. Materials Methods: pQCT was performed on male female LXRα−/−β−/−, WT at 4...

Cell-free systems derived from Xenopus eggs represent a powerful tool, intermediate between in vitro and vivo model systems. Here, we describe protocols to prepare cell-free extracts recapitulating several aspects of the DNA damage response, including damage-dependent activation ATM/ATR protein kinases checkpoint signaling pathways that inhibit initiation replication. We provide extracts, templates, kinase substrates, perform assays. In addition, related methods useful readouts response.
