The present study investigated the role of N , {′ }-dimethylbiguanide metformin (50 mg/100 g body weight in 0.05 ml water, given orally with a canulla) prevention endocrine and immune disorders provoked by hyperandrogenization dehydroepiandrosterone (DHEA) prepuberal BALB/c mice. treatment DHEA (6 0.1 oil) for 20 consecutive days, recreates mouse model that resembles some aspects human polycystic ovary syndrome (PCOS). did not modify either mass index (BMI) or blood glucose levels, but...

The present study examined the mechanism by which metformin prevents dehydroepiandrosterone (DHEA)-induced embryonic resorption in mice. Treatment with DHEA (6 mg/100 g bodyweight, 24 and 48 h post implantation) induced 88 ± 1 % diminution of both serum oestradiol (E) progesterone (P) levels. However, when (50 mg/kg bodyweight) was given together DHEA, embryo (43 3% v. 35 5% controls) E P levels were not significantly different from controls. Glucose insulin increased DHEA-treated mice but...

The aim of the present report was to study role high levels dehydroepiandrosterone (DHEA) on ovarian function and embryonic resorption during early pregnancy in BALB/c mice. Pregnant animals were injected with DHEA following both post-implantatory (DHEA-2) peri-implantatory (DHEA-6) models. Morphological studies implantation sites showed 40% DHEA-2 group while 100% observed DHEA-6 group. Serum samples groups higher estradiol a lower progesterone concentration than those control groups....

Context One of the main problems porcine in vitro maturation (IVM) is incomplete cytoplasmatic maturation. Nuclear and cytoplasmic will determine future success fertilisation embryo development. Insulin–transferrin–selenium (ITS) has insulin-like antioxidant effects, metformin (M) an insulin-sensitiser drug. Aims To assess effects adding ITS and/or M IVM media on early Methods Cumulus-oocyte complexes (COC) were with (10−4M), (0.1% v/v), + or no (Control). Key results increased glucose...

Corpus luteum regression has been described in terms of: (i) functional luteolysis - a reversible decline serum progesterone concentration; and (ii) structural irreversible morphological changes tissue remodelling events within the cellular membrane. In rats, PGF(2alpha) interleukin 1beta (IL-1beta) are involved luteolysis, by increasing ovarian lipid peroxidation, IL-1beta reducing concentrations. The aim of present report was to determine whether an early action is able regulate...

Abstract The aim of this study was to assess the presence and distribution apoptosis in porcine cumulus‐oocyte complexes (COCs) its relations with COC morphology developmental competence. COCs were obtained from slaughterhouse ovaries, classified into A1 (top category), A2, B1, B2, C, D based on their morphology. A1, B1 matured fertilized vitro, blastocyst rate compared among them. Before after vitro maturation (IVM), annexin‐V staining, terminal deoxynucleotidyl transferase dUTP nick end...

The use of vesicles co-incubated with plasmids showed to improve the efficiency cytoplasmic injection transgenes in cattle. Here, this technique was tested as a simplified alternative for delivery porcine zygotes. To aim, plasmid alone compared both diploid parthenogenic and IVF pcx-egfp injected circular (CP) at 3, 30 300 ng/μl linear (LP) ng/μl. experimental groups using parthenogenetic zygotes were follows: CP naked 3 (N = 105), 95) 65); Sham 105); control not 223); LP 78); 115) 59). The aim of this work was to study the effects dehydroepiandrosterone (DHEA) and metformin (M) on nitric oxide (NO) system oxidative stress in embryo implantation sites early pregnant mice. The biguanide M is used for treating polycystic ovary syndrome but its complete mechanism action remains unknown.Nitric has important protective roles during pregnancy, keeping uterine relaxation vascular function. However, overproduction leads nitrative by producing reactive nitrogen species. Here we...

Oocyte in vitro maturation (IVM) is crucial for subsequent embryo production. It involves acquisition of competence fertilization and development. Therefore, its optimization could have a direct impact on Dimethyl sulfoxide (DMSO) commonly used as solvent or vehicle, but also increases the membrane permeability behaves scavenger cytotoxic free radicals.
