糖類による凍結Hbの変性沈殿量,誘導形態,酸素親和性の生理機能に及ぼす影響を試験した.凝固沈殿量はシークロース,グリセロール,トレハロースとも0.4%以上の添加で,凍結期間7ヵ月の長期においても,認められなかった.0.2%添加で,わずかに沈殿が認められたが,糖無添加の3.6%に比べて著しく低かった.メト化抑制効果はシュクロース,グリセロールでほぼ同等であった.その効果は糖濃度と正の相関であった.7カ月の長期冷凍において,metHbの生成量は無添加で51%,1.6%添加で8%以下,6%添加で4%以下であった.トレハロースは1.6~3%添加のとき,metHbの生成量が最小になった.冷凍によって,糖濃度が低いほどHbの酸素平衡曲線はシグモイド型から双曲線型へ移行した.シェクロース,グリセロール,トレハロース添加によりメト化が抑制されたHbのHill定数は新鮮Hbの値3に保持され,生理機能の安定性が認められた.無添加でのHill定数は7カ月間で約1.4に低下し,P50は4.2Torrとなり,低pO2下での酸素親和性の増加が認められた.糖による安定化の機構は糖がペプチド表面の水分子と選択的...
