Abstract Hepatocellular carcinoma (HCC) is a major cause of cancer‐related mortality, while the hepatocyte mechanisms driving oncogenesis remains poorly understood. In this study, single‐nucleus RNA sequencing samples from 22 HCC patients revealed 10 distinct subtypes, including beneficial Hep0, predominantly malignant Hep2, and immunosuppressive Hep9. These subtypes were strongly associated with patient prognosis, confirmed in TCGA‐LIHC Fudan cohorts through composition deconvolution. A...

DNA methylation is essential for a wide variety of biological processes, yet the development highly efficient and robust technology remains challenge routine single-cell analysis. We developed multiplex scalable reduced representation bisulfite sequencing (msRRBS) technology. It allows cell-specific barcoded fragments individual cells to be pooled before conversion, free enzymatic modification or physical capture ends, achieves read mapping rates 62.5 ± 3.9%, covering 60.0 1.4% CpG islands...

ABSTRACT DNA methylation is essential for a wide variety of biological processes, yet the development highly efficient and robust technology remains challenge routine single-cell analysis. We developed multiplex scalable reduced representation bisulfite sequencing (msRRBS) with off-the-shelf reagents equipment. It allows cell-specific barcoded fragments individual cells to be pooled before conversion, free enzymatic modification or physical capture ends, achieves unparalleled read mapping...
