Current therapies for metastatic osseous disease frequently fail to provide a durable treatment response. To date, there are only limited therapeutic options prostate cancer, the mechanisms that drive survival of metastasis-initiating cells poorly characterized, and reliable prognostic markers missing. A high aldehyde dehydrogenase (ALDH) activity has been long considered marker cancer stem (CSC). Our study characterized differential role ALDH1A1 ALDH1A3 genes as regulators progression growth.

Erythropoiesis is a complex process driving the production of red blood cells. During homeostasis, adult erythropoiesis takes place in bone marrow and tightly controlled by erythropoietin (EPO), central hormone mainly produced renal EPO-producing The expression EPO strictly regulated local changes oxygen partial pressure (pO2) as under deprived (hypoxia) transcription factor Hypoxia Inducible Factor-2 induces EPO. However, regulation extends beyond well-established HIF-EPO axis, involves...

Abstract Cancer stem cells (CSC) are characterized by high self-renewal capacity, tumor-initiating potential, and therapy resistance. Aldehyde dehydrogenase (ALDH)+ cell population serves as an indicator of prostate CSCs with increased resistance, enhanced DNA double-strand break repair, activated epithelial-mesenchymal transition (EMT) migration. Numerous ALDH genes contribute to enzymatic activity; however, only some them showed clinical relevance. We found that ALDH1A1 ALDH1A3...

Although new targeted therapies, such as ibrutinib and idelalisib, have made a large impact on non-Hodgkin's lymphoma (NHL) patients, the disease is often fatal because patients are initially resistant to these or they eventually develop resistance. New drugs treatments necessary for patients. One attractive approach inhibit multiple parallel pathways that drive growth of hematologic tumors, possibly prolonging duration response reducing Early clinical trials tested this by dosing two in...

Abstract Endoglin (Eng) is an accessory receptor for transforming growth factor-β (TGF-β) that critical maintaining vascular integrity. Mutations in Eng cause hereditary hemorrhagic telangiectasia (HHT), resulting arteriovenous malformations (AVMs) and blood abnormalities. Despite the known association between deficiency AVMs, underlying mechanisms are unclear. In addition, role of bone marrow (BM), a major source immune cells, endothelial (EC-Eng) unexplored. We show BM vessels...

Solid tumors are typically associated with unbridled proliferation of malignant cells, accompanied by an immature and dysfunctional tumor-associated vascular network. Consequent impairment in transport nutrients oxygen eventually leads to a hypoxic environment wherein cells must adapt survive overcome these stresses. Hypoxia inducible factors (HIFs) central transcription the hypoxia response drive expression vast number survival genes cancer tumor microenvironment. HIFs tightly controlled...

Saxitoxin and gonyautoxin 2 3 are among the most toxic components of Paralytic Shellfish Poison from red tides.Being small molecules, they often require to be immobilized in order handled experimentally. Here is presented a methodology for covalently binding toxins carboxilatemodified surfaces. Both were successfully bound magnetic beads saxitoxin was additionally modified golden surface perform plasmon resonance analysis.Success evaluated through standard immune-based toxin assay.Despite...
