Significance The sphingolipid backbone ceramide is the major lipid species in stratum corneum and plays a pivotal function skin permeability barrier formation. Acylceramide an important epidermis-specific species. However, details of acylceramide production, including its synthetic genes, reactions their orders, intracellular site for have remained unclear. In present study, we identified cytochrome P450, family 4, subfamily F, polypeptide 22 (CYP4F22) as missing fatty acid ω-hydroxylase...
Abstract The incidence of dementia, a clinical symptom characterized by severe cognitive decline, is increasing worldwide. Predictive biomarkers are therefore required for early identification and management. D-amino acids in the brain contribute to function suggested as useful diagnosing dementia risk. To clarify their relationship with human we developed an method chiral metabolomics detecting slight differences amino acid amounts. Chiral tandem liquid chromatography-tandem mass...
Abstract Background Specific species of ceramides (Cer), major constituents lipids in the stratum corneum (SC), are decreased and correlated with SC barrier water-holding functions skin patients atopic dermatitis (AD) or psoriasis (Pso). However, possible correlations between Cer subclass ratios properties barrier-disrupted healthy remain unclear. The objective this study was to identify a new marker evaluate epidermal differentiation not only but also skin. Methods control subjects AD Pso...
Abstract Background The global prevalence of dementia is significantly increasing. Early detection and prevention strategies, particularly for mild cognitive impairment (MCI), are crucial but currently hindered by the lack established biomarkers. Here, we aimed to develop a high-precision screening method MCI combining D-amino acid profiles from peripheral blood samples with non-invasive subject information using nonlinear machine learning algorithms Methods A cross-sectional study was...
Abstract Coffee polyphenols (CPPs), including chlorogenic acid, exert various physiological activities. The purpose of this study was to investigate the effects CPPs on skin properties and microcirculatory function in humans. In double-blind, placebo-controlled study, 49 female subjects with mildly xerotic received either a test beverage containing (270 mg/100 mL/day) or placebo for 8 weeks. ingestion significantly lowered clinical scores dryness, decreased transepidermal water loss, surface...
Ceramides (CERs) play key roles in signal transduction and cell regulation, probably during the keratinization of human hair. Current methods using mass spectrometry (MS), however, are not sufficient to allow comprehensive analysis CER molecules, including isobaric isomeric CERs. Therefore, a method for profiling CERs was developed. The developed is based on reversed-phase liquid chromatography (RPLC) coupled atmospheric pressure chemical ionization (APCI)-MS. Comprehensive identification...
<i>Background:</i> Although it is well known that chemical oxidation with alkaline hydrogen peroxide damages human hair, little about the effects of bleaching during daily life activities which may alter hair components. <i>Objective:</i> To determine combined weathering on lipids and amino acids in hair. <i>Methods:</i> Levels were analyzed by chromatography scalp fibers collected from three different females experimentally treated...
Since ceramides (CERs) play roles in signal transduction and cell regulation, CERs of human hair might be responsible for apoptosis during keratinization, addition to their structural barrier water-holding functions. Although, we previously developed a method comprehensive profiling the hair, that was too insensitive quantitatively characterize small amount samples. The aim this study develop novel highly sensitive determination diverse CERs. is negative ion electrospray ionization mass...
A technology for the deposition of a persistent hydrophobicity to alkaline-color-treated weathered hair surfaces using 18-MEA (18-methyleicosanoic acid) is presented. Two approaches were examined in order make bind tightly surface. One was apply as an acid form and other salt or complex. It found that combination with specific cationic surfactants [stearoxypropyldimethylamine (SPDA) docosyldimethylamine (DSDA)] makes surface hydrophobic its maintained even after shampooing. Characterization...
Ethnic and racial differences in infant skin have not been well characterized. The purpose of this study was to establish whether there are ethnic similarities the stratum corneum (SC) functions Thai Chinese infants.Healthy infants 6 24 months age (N = 60; 30 Thai, Chinese) who resided Bangkok, Thailand, were enrolled. Transepidermal water loss (TEWL) SC hydration (capacitance) on thigh, buttock, upper arm measured. Ceramide content determined arm.SC remarkably different between two...
Hair lipid images, as visualized by argon sputter etching-scanning electron microscopy (ASE-SEM), reveal convex structures with a stitch pattern (SP) at the cell membrane complex (CMC) in transverse hair plane. Based on interindividual variation, different features of SP were classified into Types 0 to 4 corresponding scores 4. Observations using fibers collected from 27 Japanese females revealed significant positive correlations between and levels exogenous lipids, which suggests that...
The effect of the anteiso-branch moiety 18-MEA (18-methyleicosanic acid) to create a persistent hydrophobicity alkaline-color-treated weathered hair treated with 18-MEA/SPDA (stearoxypropyldimethylamine) was investigated by comparing straight-chain fatty acid (n-heneicosanoic acid, n-HEA) and an iso-branch (19-methyleicosanic 19-MEA) (18-MEA), using dynamic contact angle measurements, quantification LC/MS, temperature controlled atomic force microscopy (AFM). measurements indicated that is...

This study aimed to compare urinary D-amino acid levels between control and CKD-afflicted cats as a novel noninvasive method for assessing CKD. Cats were divided into CKD stage II groups accordance with the International Renal Interest Society guidelines. The DL-amino of analyzed using chiral tandem liquid chromatography-tandem mass spectrometry, their medical records investigated. group had considerably lower concentrations...
Accepted for publication February 20, 2008 Synopsis The variation of hair curvature in Japanese women was quantitatively investigated and the structure curved characterized with transmission electron microscopy (TEM) amino acid analysis. Two hundred thirty volunteers, aged from 10 to 70 years, were randomly selected. evaluation volunteers’ natural shape showed that 53% have straight hair, while remaining 47% (varying a slightly wavy frizzy style). average curl radius determined be 4.4 ± 2.3...
