In lupus, TLR7 and TLR9 mediate loss of tolerance to RNA DNA, respectively. Yet, promotes while protects from disease, implying differences in signaling. To dissect this 'TLR paradox', we generated two point mutants—lacking either ligand (TLR9K51E) or MyD88 (TLR9P915H) binding—in lupus-prone MRL/lpr mice. Ameliorated disease Tlr9K51E mice compared Tlr9-/- controls revealed a 'scaffold' protective function that is ligand- MyD88-independent. Unexpectedly,...
Invasive fungal infections can cause serious complications after lung transplant; therefore, prophylaxis with posaconazole is common. The delayed-release (DR) tablet preferred. Although the package insert states DR tablets cannot be crushed, recent data suggest it reasonable. We hypothesized that crushed could reach therapeutic levels in transplant recipients.
Introduction: Global epidemiological trends suggest that inflammatory bowel disease (IBD) is associated with industrialization but few studies have specifically evaluated urban exposure in early life. Our aim was to evaluate the impact of population density place birth on phenotype and severity IBD. Methods: From 2010-2014, IBD patients were enrolled a prospective cohort study at single center serving major metropolitan area large Hispanic population. Patients completed questionnaire...
Introduction: Genome-wide association studies have identified 163 susceptibility loci for inflammatory bowel disease (IBD) in Caucasian corhorts. Our aim was to evaluate ATG16L1, IL23R, and NOD2 variants a largely Hispanic cohort the U.S.Table 1: Minor Allele Frequencies (MAFs)Methods: IBD patients ethnically-matched controls were recruited 1:1 ratio at single center major US metropolitan with large population. Genomic DNA isolated following nucleotide polymorphisms (SNPs) analyzed: ATG16L1...

Background
Toll like receptors (TLR) 7 and 9, endosomal sensors for ssRNA dsDNA, are key mediators of lupus autoreactivity. Although considered homologous, they have opposing effects on severity: TLR7 exacerbates disease while TLR9 protects. One theory is that induces protection by competing with trafficking, thus restraining expression signaling. However, since both ligands available in lupus, TLRs supposed to signal via MyD88, this doesn't explain the differential two disease. To...
