<h3>Background and aims:</h3> The α2-gliadin-33mer has been shown to be important in the pathogenesis of coeliac disease. We aimed study mechanisms its epithelial translocation processing respect transcytotic paracellular pathways. <h3>Methods:</h3> Transepithelial passage a fluorescence-labelled was studied Caco-2 cells by using reverse-phase high-performance liquid chromatography, mass spectrometry, confocal laser scanning microscopy (LSM) fluorescence activated cell sorting...

Integrity of colon epithelium is crucial importance and, as small defects occur constantly, rapid repair (restitution) essential. To investigate the mechanism restitution, single-cell lesions were induced in mouse colonic surface epithelia by iontophoretic injection Ca2+. Closure resulting was monitored using confocal laser scanning microscopy (CLSM), and functional sealing electrophysiological techniques. Restitution evaluated time constant tau exponential decrease conductance an leak...

Enterococcus faecium NCIMB 10415 reduces diarrhoea incidence and duration in animals human study subjects. We tested whether the strain is also capable of reducing chronic gut inflammation aimed to identify mechanisms that are involved possible probiotic effects. To health-promoting strain, we used interleukin-10-deficient mice spontaneously develop fed these a diet containing for 3, 8 24 weeks, respectively. Control were which was identically composed but did not contain strain. After 3...

It was recently shown that variations in the ratio of dietary fermentable carbohydrates (fCHO) and protein (fCP) differentially affect large intestinal microbial ecology mucosal response. Here we investigated use mass spectrometry to profile changes metabolite composition colon urine associated with variation fCHO fCP physiology. Thirty-two weaned piglets were fed 4 diets a 2 × factorial design low fCHO, high fCHO. After 21 23 d, all pigs euthanized digesta profiles obtained by spectrometry....

Fluorescent dyes (e.g., dansyl, fluoresceine isothiocyanate, or naphthalimide groups) are widely used as markers to study biological properties of drugs. In order evaluate possible mediated cytotoxicity, we attached three molecules each 1,3,5-tris(3-propylamino)benzene initially synthesized core molecule for the design dendrimers. Cytotoxic effects were only observed NO2-substituted conjugate. The intracellular distribution was visualized via confocal fluorescence microscopy and pointed an...

Abstract A comprehensive data‐set from a multidisciplinary feeding experiment with the probiotic Enterococcus faecium was analyzed to elucidate effects of on growing piglets. Sixty‐two piglets were randomly assigned control (no treatment) and treatment group ( E. supplementation). Piglets weaned at 26 d. Age‐matched sacrificed for collection tissue samples 12, 26, 34 54 In addition zootechnical data, composition activity intestinal microbiota, immune cell types, responses determined. Our...
