目的研究转化生长因子-β1(TGF-β1)和IL-6在野百合碱诱导的肺动脉高压(PAH)大鼠肺组织中的表达水平及意义。 方法22只雄性SD大鼠随机分为正常对照组(n=6)、野百合碱-3周组(MCT-3w组, n=8)和MCT-5w组(n=8),皮下注射野百合碱复制肺动脉高压模型。检测右心室收缩压(RVSP)和右心室肥厚指数(RVHI),左肺组织经固定包埋切片后行van Geison染色,光镜下观察肺内小动脉病理改变,计算肺内小动脉血管壁厚度占血管直径的百分比(WT%)和血管壁面积占小动脉总横截面积的百分比(WA%)。蛋白免疫印迹法检测右肺组织TGF-β1表达水平,ELISA检测肺组织中IL-6表达水平。 结果与正常对照组相比,MCT-3w组和MCT-5w组大鼠的RVSP和RVHI均升高(P 结论TGF-β1和IL-6在野百合碱诱导的肺动脉高压大鼠肺组织中的表达水平均增高,TGF-β1和IL-6可能参与了PAH的发病和进展。
