Tumor hypoxia and aerobic glycolysis are well-known resistance factors for anticancer therapies. Here, we demonstrate that tumor-associated macrophages (TAM) enhance tumor in mice subcutaneous tumors patients with non-small cell lung cancer (NSCLC). We found a strong correlation between CD68 TAM immunostaining PET 18fluoro-deoxyglucose (FDG) uptake 98 matched of NSCLC. also observed significant glycolytic gene signatures 513 NSCLC from The Cancer Genome Atlas database. secreted TNFα to...

Significance Here, we are reporting our findings that hypoxia-inducible factor 1 (HIF-1) activation in monocytes promotes neovascularization matrigel and improves blood flow hindlimb ischemia through production of vascular endothelial growth S100A8. We found HIF-1 regulates S100A8 expression specifically isolated from unique strain transgenic mice targeting HIF pathways.

ABSTRACT Inflammatory bowel disease (IBD) is a chronic inflammatory disease, in which the intestinal epithelium loses its barrier function. Given existence of oxygen gradient and that inflammation further contributes to tissue hypoxia, we investigated role hypoxia-inducible factor (HIF), transcription activated under hypoxic conditions myeloid cells, progression IBD. To do this, utilized myeloid-specific knockout (KO) mice targeting HIF pathways, created by Cre-loxP system with human MRP8...

Cells universally adapt to ischemic conditions by turning on a transcription factor hypoxia-inducible (HIF), in which its role is known differ widely across many different types of cells. Given that microglia have been reported as an essential mediator neuroinflammation brain diseases, we examined the HIF progression acute phase stroke challenging our novel strains myeloid-specific Hif-1α or Hif-2α knockout (KO) mice created Cre-loxP system via middle cerebral artery occlusion (MCAO). We...

The cell surface transmembrane receptor TM4SF5 has been implicated in hepatocellular carcinoma (HCC), but its candidacy as a therapeutic target not evaluated. Building on findings that immunization with peptide vaccine targeting human can exert prophylactic and effects murine model of HCC, we developed monoclonal antibody to characterize expression HCC function there an anticancer strategy. We found the modulated signaling cells vitro, reducing motility, modulating E-cadherin expression,...

Microglia are brain resident macrophages rapidly responding to various stimuli exert appropriate inflammatory responses. Although they have recently been exploited as an attractive candidate for imaging neuroinflammation, it is still difficult visualize them at the cellular and molecular levels. Here we imaged activated microglia by establishing intracranial window chamber (ICW) in a mouse model of focal cerebral ischemia using two-photon microscopy (TPM), vivo. Intravenous injection...

Colon cancer is one of the most common death-related cancers in world. For treating colon cancer, it crucial to detect and remove malignant lesions early. Here, we developed hyaluronate (HA)-peanut agglutinin (PNA) conjugates for bioimaging cancer. The HA-PNA were successfully synthesized by coupling reaction between aldehyde-modified HA N-terminal amine group PNA. diagnostic imaging, rhodamine B (RhoB) was chemically conjugated onto PNA conjugates. After intraluminal injection HA-PNA-RhoB...

Pyrrolidine dithiocarbamate (PDTC) is a stable anti-oxidant or pro-oxidant, depending on the situation, and it widely used to inhibit activation of NF-kappaB. We recently reported that PDTC activates MIP-2 gene in NF-kappaB-independent c-Jun-dependent manner macrophage cells. In this work, we found signal transduction pathways mouse ES Among three different mitogen-activated protein kinase (MAPK) pathways, including extracellular-signal-regulated (ERK), p38 MAP kinase, stress-activated...

Nonalcoholic steatohepatitis (NASH) is the most prevalent chronic liver disease worldwide and increases risk for hepatocellular carcinoma (HCC) in developed countries. However, no medications have been approved to protect against NASH-driven HCC. Dapagliflozin, a sodium glucose cotransporter-2 (SGLT-2) inhibitor, improves hyperglycemia by promoting glycosuria. We investigated whether dapagliflozin prevents HCC using mouse model. Diethylnitrosamine (25 mg/kg) was intraperitoneally injected...

Abstract Myeloid cells (cells that give rise to monocytes and macrophages) are a critical component in the solid tumor microenvironment, promoting angiogenesis recurrence therapies. Recent literature have extensively demonstrated hypoxia-inducible factor (HIF) is key transcription myeloid responding hypoxic stimuli including tumors inflammation by secreting various cytokines growth factors. Although number of studies reported hypoxia exposure primary macrophages induces HIF-1 activation...
