Neurofibromatosis 2 (NF2) is a tumor suppressor, although the molecular mechanism accounting for this effect remains unknown. Here, we show that merlin exerts its activity by inhibiting phosphatidylinositol 3-kinase (PI3-kinase), through binding to PIKE-L. Wild-type merlin, but not patient-derived mutant (L64P), binds PIKE-L and inhibits PI3-kinase activity. This suppression of results from disrupting PI3-kinase. In addition, as well schwannoma cell growth abrogated single point mutation...

Protein engineering is a powerful tool for improving the properties of enzymes. However, large changes in enzyme are still challenging traditional evolution strategies because they usually require multiple amino acid substitutions. In this study, feasible approach by combination fragment swapping and semi-rational design was developed nitrilase. A chimera BaNIT harboring 12 substitutions obtained using nitrilase from Arabis alpine (AaNIT) Brassica rapa (BrNIT) as parent enzymes, which...

Abstract The NF2 tumor suppressor gene encodes an intracellular membrane-associated protein, called merlin, which belongs to the band 4.1 family of cytoskeleton-associated proteins that link cell surface glycoproteins actin cytoskeleton. Merlin suppresses phosphatidylinositol 3-kinase (PI3K)/Akt signaling by directly binding and inhibiting stimulatory activity PIKE-L on PI3K. Akt feeds back phosphorylates merlin provokes its polyubiquitination degradation. Here, we show phosphorylation...
