The enzymatic carboligation of aldehydes (C–C bond formation) catalyzed by benzaldehyde lyase (BAL) affords chiral α-hydroxy-ketones under mild reaction conditions in aqueous media. To enhance substrate and product availability conditions, processes are often set-up using either DMSO as co-solvent, or MTBE second organic phase. Although efficient, leads to difficulties separation during downstream processing, with wastewater formation. provides a cleaner straightforward work-up, but its...
The aims were to isolate a raw starch-degrading α-amylase gene baqA from Bacillus aquimaris MKSC 6.2, and characterize the product through in silico study its expression Escherichia coli.A 1539 complete open reading frame of B. 6·2 has been determined by employing PCR inverse techniques. Bioinformatics analysis revealed that 6.2 (BaqA) no starch-binding domain, together with few putative α-amylases bacilli may establish novel GH13 subfamily most closely related GH13_1. Two consecutive...
A one-pot multi-step approach comprising enzymatic oxidation-hydroxymethylation-reduction enables the synthesis of optically active α-aryl vicinal diols with high yields and enantioselectivities. Formaldehyde required for hydroxymethylation step is enzymatically produced in situ using less hazardous methanol as substrate.
Abstract α-Amylases are glycoside hydrolase enzymes that act on the α(1→4) glycosidic linkages in glycogen, starch, and related α-glucans, ubiquitously present Nature. Most α-amylases have been classified family 13 with a typical (β/α) 8 -barrel containing two aspartic acid one glutamic residue play an essential role catalysis. An atypical α-amylase (BmaN1) only of three invariant catalytic residues was isolated from Bacillus megaterium strain NL3, bacterial isolate sea anemone Kakaban...
Abstract Partially purified α‐amylase from Bacillus aquimaris MKSC 6.2, a bacterium isolated soft coral Sinularia sp., Merak Kecil Island, West Java, Indonesia, showed an ability to degrade raw corn, rice, sago, cassava, and potato starches with adsorption percentage in the range of 65–93%. Corn has highest degree hydrolysis followed by cassaca, sago consecutively. The end products starch were mixture maltose, maltotriose, maltotetraose, maltopentaose, maltohexaose, small amount glucose.
The severe acute respiratory syndrome coronavirus 2 -related global COVID-19 pandemic has been impacting millions of people since its outbreak in 2020. vaccination proven highly efficient reducing illness severity and preventing infection-related fatalities. World Health Organization granted emergency use approval to multiple, including protein subunit technology-based, vaccines. Foreseeably, additional vaccine development would be essential meet the accessible growing demand for effective...
A land-locked marine lake Kakaban with its significant ecological paramaters provides a unique habitat for bacteria novel biotechnology potential that uses diverse array of catalytic agents, including α-amylase. Aiming at the isolation raw starch degrading α-amylase from biodiversity, gene encoding BmaN2 sea anemone associated bacterium Bacillus megaterium NL3 was cloned and expressed in Escherichia coli ArcticExpress (DE3). It comprises an open reading frame 1,563 nucleotides 520 amino...
The aggregation of recombinant proteins into inclusion bodies is a major problem for expression in bacterial systems. must be solubilized and the denatured protein renatured if an active molecule to recovered. We have developed such procedure N-terminal domain tissue inhibitor metalloproteinases-2 [TIMP-2-(1-127)], small mammalian containing three disulfide bonds. Conditions its renaturation were determined by studying refolding behaviour reduced mammalian-cell-expressed TIMP-(1-127)...
Nanofibrous membrane has a potential to use in filtration technology with electrospinning as one of the techniques used synthesizing nanofibers. Polyacrylonitrile (PAN) nanofibrous membranes various fibers diameters were electrospun by varying its precursor solution concentration. The average PAN obtained from concentrations 6, 9, 12, and 14 wt% 341, 534, 1274, 2107 nm, respectively. Filtration media for apple juice clarification bilayer-structured made on commercial cellulose microfibrous...
Salmonella typhimurium, that cause typhoid fever in mice was used as a model to study humans. This is aimed determine the immunogenicity of recombinant protein Fim-C-S. typhimurium and specificity antibody anti-recombinant ddY mice. The studies by ELISA indicated evokes strong immune response mice, particularly experiment group. evaluated Western Immunobloting technique can detect 31 kDa an antigen. data reported immunogenic specific. results this be develop vaccine candidate against typhoid.
Mutations in katG gene are often associated with isoniazid (INH) resistance Mycobacterium tuberculosis strain. This research was perfomed to identify the mutation clinical isolate (L8) that is resistant INH at 1 μg/ml. In addition characterize catalase-peroxidase of KatG L8 and perform ab initio structural study protein get a more complete understanding drug activation resistance mechanism. The cloned expressed Escherichia coli, then followed by characterization KatG. structure modelling...
In this work, the amorphous HKUST-1 nanoparticles (aHKUST1) and copper hydroxide nanosheet (Cu(OH) 2 NS) were successfully prepared by involving diethanolamine (DEOA) applied as an immunosensor to detect dengue virus (DENV-3) NS1. 35 wt% of DEOA was found not only provide a base environment but also act crystal modifier for HKUST-1. The changes crystallinity morphology into highly uniform nanoparticles, respectively. addition, Cu(OH) NS obtained immersing aHKUST1 NPs in water 6 h....
Most of isoniazid-resistant Mycobacterium tuberculosis evolved due to mutation in the katG gene encoding catalase-peroxidase. A set new mutations, namely T1310C, G1388T, G1481A, T1553C, and A1660G, which correspond amino acid substitutions L437P, R463L, G494D, I518T, K554E, L10 clinical isolate M. was identified. The wild-type mutant KatG proteins were expressed Escherichia coli BL21(DE3) as a protein 80 kDa based on sodium dodecyl sulphate-polyacrylamide gel electrophoresis analysis....
Fructose and Fructo-Oligosaccharides (FOS) are derived more practical from the enzymatic hydrolysis reaction of inulin. In this study, gene fragments that encodes inulin enzyme were isolated genomic B. licheniformis by Polymerase Chain Reaction (PCR) technique using new primers designed on conserved domain in family GH32 enzymes.Gene cloned into pGEM-T vector with E. coli as host cells determined sequence nucleotide bases.Size fragment have been found 539 bp DPE.slF DPE.eR primer pair. The 179...
Recombinant small hepatitis B surface antigen (sHBsAg) is used as a vaccine component to prevent virus infection. As an attempt produce local recombinant sHBsAg, PCR-amplified DNA fragment encoding Indonesia sHBsAg which belongs genotype and adw2 subtype was cloned into Hansenula polymorpha expression vector pHIPX4 by using recombination method. The resulted pHIPX4-sHBsAg integrated the alcohol oxidase (AOX) locus of H. NCYC495 genome regulated under control AOX promoter. carrying coding...
