A series of pyrazine and quinoxaline derivatives have been synthesized, their activity against M. tuberculosis (Mtb) Mycobacterium avium (MAC) are reported. The 4-acetoxybenzyl ester pyrazinoic acid 4‘-acetoxybenzyl 2-quinoxalinecarboxylate showed excellent Mtb (MIC ranges less than 1−6.25 μg/mL) but only modest MAC (MICs 4−32 μg/mL).
ABSTRACT This study extends earlier reports regarding the in vitro and vivo efficacies of nitroimidazopyran PA-824 against Mycobacterium tuberculosis . was tested a broad panel multidrug-resistant clinical isolates found to be highly active all (MIC < 1 μg/ml). The activity M. also assessed grown under conditions oxygen depletion. showed significant at 2, 10, 50 μg/ml, similar that metronidazole, dose-dependent manner. In short-course mouse infection model, efficacy 50, 100, 300 mg/kg...
ABSTRACT Many of the current antimycobacterial agents require some form cellular activation unmasking reactive groups, which in turn will bind to their specific targets. Therefore, understanding mechanisms antimycobacterials not only helps decipher drug resistance but may also facilitate development alternative strategies or analogues that do such processes. Herein, through use genetically defined strains Mycobacterium bovis BCG we provide evidence EthA, previously shown activate...
Analyses of soil water and groundwater samples from a high-elevation coniferous ecosystem in New England indicate that sulfate anions supply 76 percent the electrical charge balance leaching solution. This result implies atmospheric inputs sulfuric acid provide dominant source both H(+) for cation replacement mobile transport subalpine soils northeastern region affected by precipitation. In relatively unpolluted regions, carbonic organic acids dominate processes.
On the order of hundreds absorption, distribution, metabolism, excretion, and toxicity (ADME/Tox) models have been described in literature past decade which are more often than not inaccessible to anyone but their authors. Public accessibility is also an issue with computational for bioactivity, ability share such still remains a major challenge limiting drug discovery. We describe creation reference implementation Bayesian model-building software module, we released as open source component...
Tuberculosis is a global health dilemma. In 2016, the WHO reported 10.4 million incidences and 1.7 deaths. The need to develop new treatments for those infected with Mycobacterium tuberculosis ( Mtb) has led many large-scale phenotypic screens thousands of active compounds identified in vitro. However, limited funding, efforts discover molecules against Mtb needs be more efficient. Several computational machine learning approaches have been shown good enrichment hit rates. We curated small...
The Southern Alps are developing as a consequence of oblique collision between the Pacific and Australian plates. lie on west side South Island New Zealand create massive rain shadow where greater than 12 m/year falls coast semiarid conditions exist to east. rain-out effect across mountains causes precipitation have δD δ18O values averaging −30‰ −5.5‰, whereas in east is isotopically lighter (δD=−72‰ δ18O=−9.8‰). Such large differences isotopic composition would not existed prior development...
BackgroundMycolic acids are a complex mixture of branched, long-chain fatty acids, representing key components the highly hydrophobic mycobacterial cell wall. Pathogenic mycobacteria carry mycolic acid sub-types that contain cyclopropane rings. Double bonds at specific sites on precursors modified by action synthases (CMASs). The latter belong to family S-adenosyl-methionine-dependent methyl transferases, which several have been well studied in Mycobacterium tuberculosis, namely, MmaA1...
ABSTRACT FtsZ, a bacterial homolog of tubulin, forms structural element called the FtsZ ring (Z ring) at predivisional midcell site and sets up scaffold for assembly other cell division proteins. The genetic aspects FtsZ-catalyzed its dynamics in Mycobacterium tuberculosis are unknown. Here, with an M. strain containing TB tagged green fluorescent protein as sole source we examined structures under various growth conditions. We found that Z rings present approximately 11% actively growing...
