Mitochondrial production of oxidants contributes to a variety pathological conditions including the vascular complications diabetes, neurodegenerative diseases, and cellular senescence. We postulated that transcriptional coactivator, peroxisome proliferator activated receptor-gamma coactivator 1alpha (PGC-1alpha), major regulator oxidative metabolism mitochondrial biogenesis, could be involved in regulation antioxidant defense system endothelial cells. We show PGC-1alpha is present human,...

Abstract Introduction The mammalian adult heart maintains a continuous, low cardiomyocyte turnover rate throughout life. Although many cardiac stem cell populations have been studied, the natural source for homeostatic repair has not yet defined. Polycomb protein BMI1 is most representative marker of mouse systems. We evaluated relevance and role Bmi1 + cells in physiological homeostasis. Methods CreER/+ ; Rosa26 YFP/+ ( -YFP) mice were used lineage tracing strategy. After tamoxifen (TM)...

ABSTRACT Number of molecules and Brightness (N&B) has been proposed for measuring the molecular brightness number fluorophores in time‐sequence images, live cells. If fluorescently tagged‐proteins are mobile illumination volume, stoichiometry their oligomers can be derived from increase fluorescent dyes due to clustering. We examine aspects concerning extra‐fluctuation effects induced by cell shifts photobleaching, which yield large overestimates clusters size sub‐unit counts. develop an...

In a previous study, we demonstrated that endothelial microvesicles (eMVs) have well‐developed enzymatic team involved in reactive oxygen species detoxification. the present paper, demonstrate eMVs can synthesize reducing power (NAD(P)H) nourishes this team, especially those derived from senescent human umbilical vein cells. Moreover, molecules nourish machinery NAD(P)H synthesis are blood plasma metabolites: lactate, pyruvate, glucose, glycerol, and branched‐chain amino acids. Drastic...

The fusion of bone marrow (BM) hematopoietic cells with hepatocytes to generate BM derived (BMDH) is a natural process, which enhanced in damaged tissues. However, the reprogramming needed BMDH and identity resultant essentially unknown. In mouse model chronic liver damage, here we identify modification chromatin structure nucleus during formation, accompanied by loss key transcription factor PU.1/Sfpi1 (SFFV proviral integration 1) gain hepatic transcriptional regulator HNF-1A homeobox A...

Both fibroblast growth factor-2 (FGF2) and neural cell adhesion molecule (NCAM) trigger FGF receptor 1 (FGFR1) signaling, however they induce remarkably distinct trafficking cellular responses. The molecular basis of such a dichotomy the role types ligand-receptor interactions remain elusive. Number molecules Brightness (N&B) analysis revealed that FGF2 NCAM promote different FGFR1 assembly dynamics at plasma membrane. stimulation elicits long-lasting cycles short-lived monomers...

An analysis of liver cell populations from both adult and 21 day pregnancy rat fetuses (E21) was carried out. The results show that E21 hepatocytes express OX‐43, as do endothelial cells but not hepatocytes. OX‐43 could be used in future a marker for the hepatocyte maturation.
