Research on the specificities, functions, and maturation of T cells would be greatly aided by a collection monoclonal antibodies which distinguishes different types TCR. With this end in mind hamsters were immunized tested for production pan-reactive anti-mouse alpha beta TCR antibodies. In report we describe properties uses mAb, H57-597, produced from one these animals. The mAb reacts with surface receptors all TCR-bearing does not react gamma delta+ cells. an immobilized form, antibody can...

Abstract IL-17A is originally identified as a proinflammatory cytokine that induces neutrophils. Although production by CD4+ Th17 T cells well documented, it not clear whether produced and participates in the innate immune response against infections. In present report, we demonstrate expressed liver of mice infected with Listeria monocytogenes from an early stage infection. important protective immunity at listerial infection because IL-17A-deficient showed aggravation response. The major...

Two variants of the AKR thymoma BW5147 have been isolated which can no longer express functional TCR alpha- and beta-chains. By generating hybridomas with these variant fusion lines, any normal T lymphocyte, including TCR-gamma/delta, be studied at a clonal level, without interference BW5147-derived receptor chains. In this study one has useful in identifying reactivity to allogeneic MHC Ag itself.

Abstract Murine γδ T cell subsets, defined by their Vγ chain usage, have been shown in various disease models to distinct functional roles. In this study, we examined the responses of two main peripheral Vγ1+ and Vγ4+ cells, during collagen-induced arthritis (CIA), a mouse model that shares many hallmarks with human rheumatoid arthritis. We found whereas both subsets increased number, only cells became activated. Surprisingly, these appeared be Ag selected, based on preferential Vγ4/Vδ4...

Small synthetic peptides that correspond to different portions of the 65-kilodalton mycobacterial heat shock protein (Hsp65) were used identify a putative antigenic epitope for γδ cells. Weaker responses equivalent portion autologous homolog, mouse Hsp63, also seen. The stimulatory overlaps with an recognized by arthritogenic αβ T cell clones. data suggest cells have role in autoimmune disorders and imply these recognize ligands mechanism similar lymphocytes, is, form small processed...

Inflammation-induced pulmonary fibrosis (PF) leads to irreversible loss of lung function and is a predictor mortality in numerous diseases. Why some subjects with inflammation but not others develop PF unclear. In mouse model hypersensitivity pneumonitis that progresses upon repeated exposure the ubiquitous microorganism Bacillus subtilis, γδ T cells expand inhibit collagen deposition. We show subset these represents predominant source Th17 cytokine IL-22 this model. Preventing expression...

The kinetics and order of rearrangements in the gene complex encoding T-cell-receptor beta chains were studied by Southern blot hybridization a collection hybridomas derived from fetal thymocytes at various stages ontogeny (day 14 to day 17). Our results show steady increase frequency rearranged complexes during this period suggest that these occur within thymus. beta-chain diversity region (D beta) joining (J preceded other types rearrangements. Host defenses against experimental listeriosis in mice involve neutrophils, macrophages, NK cells, and alpha beta T cells. Recently gamma delta cells have also been implicated antilisterial resistance. However, their specific role has remained unclear. Here we show that efficient resistance to infection by this bacterium depends on the functions of both primary secondary responses. We present evidence these are complementary. In livers cell-depleted mice, bacteria grow large numbers within...

Abstract Hypersensitivity pneumonitis is an environmental lung disease characterized by a diffuse mononuclear cell infiltrate in the that can progress to pulmonary fibrosis with chronic exposure inhaled Ag. Using well-established murine model of hypersensitivity pneumonitis, we repeatedly exposed C57BL/6 mice Saccharopolyspora rectivirgula investigate whether T cells are required for fibrosis. In absence αβ cells, TCRβ−/− S. 4 wk had markedly decreased infiltrates and collagen deposition...

Previously, we detected a subset of gamma delta T cells in the newborn mouse thymus that responded to mycobacterial heat shock protein Hsp60, as well with what seemed be self-antigen. All these expressed V 1, most often association 6+. It was not clear, however, whether similar, mature Hsp60 reactivity are common outside thymus, or rather, they largely eliminated during development. From data presented here, estimate responding comprise 10-20% normal splenic and lymph node cells. Such cells,...

Abstract IL‐17 is produced not only by CD4 + αβ T cells, but also CD8 NKT and γδ plus some non‐T including macrophages neutrophils. The ability of to deploy neutrophils sites inflammation imparts this cytokine with a key role in diseases several types. Surprisingly, cells are responsible for much the disease models, particularly early on.

To study the origin of and degree T-cell antigen receptor (TCR) diversity Thy-1+ dendritic epidermal cells (Thy-1+ dECs) in mice, we have developed a monoclonal antibody (mAb 536) to gamma delta TCR. mAb 536 binds stimulates interleukin 2 secretion from dEC but not that express TCR composed alpha beta chains. precipitates CD3-associated chains lysates radioiodinated dECs. Allergic airway inflammation and hyperreactivity are modulated by gammadelta T cells, but different experimental parameters can influence the effects observed. For example, in sensitized C57BL/6 BALB/c mice, transient depletion of all TCR-delta(+) cells just before challenge resulted hyperresponsiveness (AHR), caused hyporesponsiveness when initiated i.p. sensitization. Vgamma4(+) strongly suppressed AHR; their relieved suppression challenge, not sensitization, they AHR transferred into...

Abstract A specific subset of gamma delta T lymphocytes bearing a V 6V 1-encoded TCR is known to populate the normal nonpregnant murine uterine and vaginal epithelium. However, residing at maternal-fetal interface during pregnancy have not yet been investigated.
