The direct reduction of alcohols using chlorodiphenylsilane as a hydride source in the presence catalytic amount indium trichloride is described. Benzylic alcohols, secondary and tertiary were effectively reduced to give corresponding alkanes high yields. A compound bearing both primary hydroxyl groups was only at site afford alcohol after workup with Bu(4)NF. This system showed chemoselectivity for group while not reducing other functional that are readily by standard systems. Thus ester,...

Abstract Currently, there are no treatments for Alport syndrome, which is the second most commonly inherited kidney disease. Here we report development of an exon-skipping therapy using antisense-oligonucleotide (ASO) severe male X-linked syndrome (XLAS). We targeted truncating variants in exon 21 COL4A5 gene and conducted a type IV collagen α3/α4/α5 chain triple helix formation assay, vitro vivo treatment efficacy evaluation. show that skipping enabled trimer formation, leading to...

Abstract Incorporating stimuli‐responsive components into RNA constructs provides precise spatiotemporal control over structures and functions. Despite considerable advancements, the utilization of redox‐responsive stimuli for activation caged RNAs remains scarce. In this context, we present a novel strategy that leverages post‐synthetic acylation coupled with chemistry to exert RNA. To achieve this, design synthesize series acylating reagents specifically tailored introducing...

The indium(III) chloride/chlorotrimethylsilane system acts as a catalyst for the Sakurai−Hosomi reaction between allylsilanes and aldehydes. When γ-substituted such crotylsilane prenylsilane are used high regioselectivity γ-addition is observed. Smaller amounts of (5 mol %) effectively afforded allylation products, although larger (20 gave no product whatsoever. Other group 13 Lewis acids AlCl3 BF3·OEt2 did not have catalytic activity even when combined with chlorotrimethylsilane. (©...

The reaction of carbonyls and chlorodimethylsilane was effectively catalyzed by indium(III) hydroxide afforded the corresponding deoxygenative chlorination products, in which carbonyl carbon accepted two nucleophiles (H Cl) with releasing oxygen. Only In(OH)3 reaction, typical Lewis acids such as TiCl4, AlCl3, BF3.OEt2 showed no catalytic activity. mechanism this includes initial hydrosilylation followed chlorination. Acylation of RNA at 2'-OH groups is widely applied in mapping structure and recently for controlling function. Reactions are described that install the smallest 2-carbon acyl on RNA-namely, 2'-O-acetyl 2'-O-carbonate groups. Hybridization thermal melting experiments performed to assess effects duplex formation. Both reagents can be employed lower concentrations map secondary by reverse transcriptase primer extension (SHAPE) methods.

Abstract Investigations into the human 8-oxodGTPase, MutT Homolog 1 (MTH1), have risen sharply since first-in-class MTH1 inhibitors were reported to be highly tumoricidal. However, as a cancer therapeutic target is currently controversial because subsequently developed did not exhibit similar cytotoxic effects. Here, we provide first direct evidence for MTH1-independent 8-oxodGTPase function in cells and tumors, using novel ATP-releasing guanine-oxidized (ARGO) chemical probe. Our studies...
