A gene encoding a maltogenic amylase of Bacillus stearothermophilus ET1 was cloned and expressed in Escherichia coli . DNA sequence analysis indicated that the could encode 69 627‐Da protein containing 590 amino acids. The predicted acid enzyme shared 47−70 % identity with sequences from licheniformis , neopullulanase B. cyclodextrin hydrolase (CDase) I‐5 an alkalophilic strain. In addition to starch, pullulan cyclodextrin, hydrolyze isopanose, but not panose, glucose maltose. Maltogenic...

ABSTRACT A maltogenic amylase gene was cloned in Escherichia coli from a gram-negative thermophilic bacterium, Thermus strain IM6501. The encoded an enzyme (ThMA) with molecular mass of 68 kDa which expressed by the expression vector p6xHis119. optimal temperature ThMA 60°C, higher than those other amylases reported so far. Thermal inactivation kinetic analysis indicated that it stabilized presence 10 mM EDTA. harbored both hydrolysis and transglycosylation activities. It hydrolyzed...

Mutants with deletion mutations in the glg and mal gene clusters of Escherichia coli MC4100 were used to gain insight into glycogen maltodextrin metabolism. Glycogen content, molecular mass, branch chain distribution analyzed wild type ΔmalP (encoding phosphorylase), ΔmalQ amylomaltase), ΔglgA synthase), derivatives. The showed increasing amounts when grown on glucose, maltose, or maltodextrin. When strains content was 20 times higher strain (0.97 mg/mg protein) than (0.05 protein). had...

Wild yeasts on the surface of various fruits including grapes were surveyed to obtain yeast strains suitable for fermenting a novel wine with higher alcohol content and supplemented rice starch. We considered selected characteristics, such as tolerance osmotic pressure, capability utilizing maltose, starch hydrolysis.Among 637 putative isolates, 115 exhibiting better growth in yeast-peptone-dextrose broth containing 30% dextrose, 7% alcohol, or 2% maltose selected, well five α-amylase...

Biofilm-related nosocomial infections may cause a wide range of life-threatening infections. In this regard, Pseudomonas aeruginosa biofilm is becoming serious health burden due to its capability develop resistance natural and synthetic drugs. The utilization nanoparticles that inhibit formation one the major strategies control caused by biofilm-forming pathogens. Hydroxyapatite (HA) ceramic material having properties similar bones. Herein, co-precipitation method followed microwave...

The glycogen branching enzyme from Vibrio vulnificus (VvGBE) transfers short side chains (DP 3–5) significantly greater than any other bacterial (GBE). To elucidate the role of N‐domain VvGBE in unique pattern, domain‐truncated (N1 and N) N1‐domain‐swapped (with N1 replacing counter part Escherichia coli GBE) mutants were constructed. truncation synthesized branched products with a greatly reduced proportion chains. swapping mutant exhibited pattern chain region similar to that VvGBE. We...

Maltogenic amylase and α-glucanotransferase (α-GTase) were employed in an effort to develop efficient process for the production of isomaltooligosaccharides (IMOs). Bacillus stearothermophilus maltogenic (BSMA) α-GTase from Thermotoga maritima overexpressed Escherichia coli using overexpression vectors. An IMO mixture containing 58% various IMOs was produced liquefied corn syrup by hydrolyzing transglycosylation activities BSMA alone. When reacted simultaneously, content increased 68%...

The physiological functions of two amylolytic enzymes, a maltogenic amylase (MAase) encoded by yvdF and debranching enzyme (pullulanase) amyX, in the carbohydrate metabolism Bacillus subtilis 168 were investigated using yvdF, amyX mutant strains. An immunolocalization study revealed that YvdF was distributed on both sides cytoplasmic membrane periplasm during vegetative growth but cytoplasm prespores.
