Summary A new methodology for blastocyst biopsy that uses a 1.48μm in diode laser is described. Trophectoderm cells are biopsied after laster zona drilling and culture, fixed processed fluorescent situ hybridisation (FISH) analysis. Preliminary results on the efficiency of procedure recovery rate promising. Blastocyst useful tool preimplantation genetic diagnosis (PGD) as it allows more reliable widens diagnostic possibilities account higher number obtained biopsy.

Next-generation sequencing (NGS) has the capacity of carrier screening in gamete donation (GD) programs. We have developed and validated an NGS carrier-screening test (qCarrier test) that includes 200 genes associated with 368 disorders (277 autosomal recessive 37 X-linked). Carrier is performed on oocyte candidates male partner recipient. Carriers X-linked conditions are excluded from GD program, whereas donors chosen who do not carry mutations for same gene/disease as recipients. The...

Summary Shortly after the implementation of comprehensive chromosome screening (CCS) techniques for preimplantation genetic testing aneuploidies (PGT-A), discussion about transition from day 3 to blastocyst stage biopsy was initiated. Trophectoderm with CCS is meant overcome limitations cleavage-stage and single-cell analysis. The aim this study assess results obtained in our PGT-A programme new strategy. Comparisons between 179 cycles (D+3) fresh embryo transfer, 204 trophectoderm deferred...

Three cases of preimplantation genetic diagnosis (PGD) (two for sexing and one aneuploidy screening) are presented. Embryo biopsy was performed at day 3 established with fluorescent in situ hybridization (FISH). Embryos not used replacement were cultured sequential media blastocyst development. Blastocyst rate 39.3 per cent. Confirmations FISH blastocysts arrested embryos. Mosaicism observed 7/8 (mean number cells analysed: 55.5) 5/8 The percentage abnormal 17.1 cent 54 Polyploid 4/8...

BACKGROUND: Contraceptive treatment before gonadotrophin-releasing hormone agonist administration presents advantages in women with a tendency to hyper-response and simplifies donor–recipient synchronization. This study compares response gonadotrophin stimulation under hypophyseal suppression oocyte donors or without vaginal contraceptive pretreatment. METHODS: One hundred ninety were recruited single centre prospectively assigned one of two groups, according the day week menstruation...

A twin female pregnancy was obtained in a haemophilia carrier after two preimplantation diagnosis cycles. The embryonic sex of biopsied blastomeres determined with the use dual fluorescence in-situ hybridization directly labelled DNA probes specific for X and Y chromosome. confirmed by means ultrasonography amniocentesis at 14th week amenorrhoea. patient delivered healthy females Caesarian section 37th pregnancy.

The transition in biopsy timing from blastomere to trophectoderm has led a remarkable decrease the percentage of undiagnosed blastocysts. However, patients with few or no euploid blastocysts can be affected by this residual diagnosis failure. aim study is assess whether blastocyst rebiopsy and revitrification an efficient safe procedure applied cases results after analysis. Fifty-three agreed warming 61 perform second PGT-A aCGH. Only 75.4% survived, reexpanded, could rebiopsied. After...
