The use of antibiotic-coated implants may reduce the rate infection and facilitate fracture healing after surgical treatment tibial shaft fractures. A new biodegradable gentamicin-loaded coating an implant (UTN PROtect) was CE-certified in August 2005. In this prospective, non-randomized case series, we investigated clinical, laboratory radiological outcomes 21 patients who underwent closed or open fractures, as well revisions with UTN PROtect gentamicin-coated intramedullary nail.
Abstract Developmental osteogenesis, physiological bone remodelling and fracture healing require removal of matrix cellular debris. Osteoclasts generated by the fusion circulating monocytes degrade bone, whereas identity cells responsible for cartilage resorption is a long-standing controversial question. Here we show that degradation chondrocyte phagocytosis are mediated fatty acid binding protein 5-expressing representing septoclasts, which have mesenchymal origin not derived from...
Abstract Bone regeneration requires a well-orchestrated cellular and molecular response including robust vascularization recruitment of mesenchymal osteogenic cells. In femoral fractures, angiogenesis osteogenesis are closely coupled during the complex healing process. Here, we show with advanced longitudinal intravital multiphoton microscopy that early vascular sprouting is not directly to osteoprogenitor invasion calvarial bone regeneration. Early osteoprogenitors emerging from periosteum...
<h3>Objective</h3> To elucidate the mechanisms involved in cartilage damage an experimental model of rheumatoid arthritis (RA) by specifically addressing time course extracellular matrix degradation and contribution cell–matrix interactions for initiation perpetuation this process. <h3>Methods</h3> The human tumour necrosis factor (TNF) transgenic (hTNFtg) mouse RA was used to analyse pannus attachment destruction, respectively, crossed hTNFtg mice with interleukin (IL)-1−/−...
Sensory and sympathetic nerve fibers (SNF) innervate bone epiphyseal growth plate. The role of neuronal signals for proper endochondral ossification during skeletal is mostly unknown. Here, we investigated the impact absence sensory neurotransmitter substance P (SP) removal SNF on callus differentiation, a model in adult animals, formation. In order to generate callus, tibia fractures were set left hind leg wild type (WT), tachykinin 1-deficient (Tac1 −/−) mice (no SP) animals without SNF....
"Candidatus Liberibacter asiaticus" is the causative bacterium associated with citrus greening disease. "Ca transmitted by Diaphorina citri more efficiently when it acquired nymphs rather than adults. Why this occurs not known. We compared midguts of D. insects reared on healthy or asiaticus"-infected trees using quantitative PCR, confocal microscopy, and mitochondrial superoxide staining for evidence oxidative stress. Consistent its classification as propagative, titers were higher in...
Recent studies reveal an important role of vascular endothelial growth factor (VEGF)-induced angiogenesis in degenerative tendon diseases. The way how VEGF influences mechanical properties the tendons is not well understood yet. We here hypothesized that tendinopathy results a hypoxia-mediated stimulation and stability impaired angiogenic process by VEGF-induced matrix metalloproteinases (MMPs). A modified situ freezing model patellar was used to create tendinopathy. 0, 7, 14, 28 days...
Abstract Background Osteoclasts are the tissue-specific macrophage population of bone and unique in their bone-resorbing activity. Hence, they fundamental for physiology health disease. However, efficient protocols isolation study primary human osteoclasts scarce. In this study, we aimed to establish a protocol, which enables differentiation functional from monocytes. Results Human monocytes were isolated through double-density gradient donor blood. Compared standard schemes polystyrene cell...
Previous studies showed that growth factors dramatically stimulate healing processes in bone. However, the long-term effect of locally applied on fracture remains unclear. In considering safety using factors, it is necessary to elucidate after initial stimulation, stops and result a normally healed tissue. Therefore, purpose present study was investigate time course during factor (GF) stimulated unstimulated closed tibial model rats. A well established local drug delivery system used. IGF-I...
Abstract Objective Syndecan 4, a heparan sulfate proteoglycan, has been associated with osteoarthritis. The present study was undertaken to analyze the functional role of syndecan 4 in endochondral ossification mouse embryos and adult fracture repair, which, like osteoarthritis, involves an inflammatory component. Methods Sdc4 promoter activity analyzed −/− lacZ‐knockin mice, using β‐galactosidase staining. Endochondral from embryonic day 16.5 assessed by histologic immunohistologic Bone...
The overexpression of tumor necrosis factor (TNF)-α leads to systemic as well local loss bone and cartilage is also an important regulator during fracture healing. In this study, we investigate how TNF-α inhibition using a targeted monoclonal antibody affects healing in driven animal model human rheumatoid arthritis (RA) elucidate the question whether enduring anti therapy after trauma beneficial or not.A standardized femur was applied wild type transgenic mice (hTNFtg mice), which develop...
Abstract Heterotopic ossification (HO) represents a common problem after tendon injury with no effective treatment yet being developed. Tenomodulin ( Tnmd ), the best-known mature marker for lineage cells, has important effects in tissue aging and function. We have reported that loss of leads to inferior early repair characterized by fibrovascular scaring therefore hypothesized its lack will persistently cause deficient during later stages. knockout −/− ) wild-type (WT) animals were...
Abstract Growth factors IGF‐I and TGF‐β1 are known to stimulate fracture healing. The purpose of this study was investigate the role locally applied during early phase healing (Days 5, 10, 15 after fracture) on cellular processes like proliferation differentiation in a rat model. Two different immunohistochemical markers were used analyze cell proliferation: (1) injection thymidine analogue BrdU subsequent staining for BrdU‐positive nuclei, (2) antibody against “proliferating nuclear...
Wounding citrus induces the production of antifungal compound 1, isolated from peel injured grapefruit, Citrus paradisa, and orange, sinensis. Interpretation ms, uv, 1D 2D nmr spectroscopic data led to identification new as 3-[4-hydroxy,3-(3-methyl-2-butenyl)-phenyl]-2-(E)-propenal.
Huanglongbing {HLB [associated with Candidatus Liberibacter sp. (CLas)]} and asiatic citrus canker {ACC [causal organism Xanthomonas citri ssp. (XCC)]} are bacterial diseases that seriously threaten sustainability of the Florida ( Citrus sp.) industry. Sweet orange sinensis ) grapefruit paradisi highly susceptible to ACC improvement through conventional breeding is a long-term process, making transgenic solutions attractive. No strong HLB resistance has been identified within cultivated...
Abstract Bone loss is one of the consequences aging, leading to diseases such as osteoporosis and increased susceptibility fragility fractures therefore considerable morbidity mortality in humans. Here, we identify microRNA‐146a (miR‐146a) an essential epigenetic switch controlling bone with age. Mice deficient miR‐146a show regular development their skeleton. However, while WT mice start lose age, animals continue accrue throughout life span. Increased mass due generation activity...
Thirty‐six isolates of the fasciation‐inducing bacterium Rhodococcus fascians were evaluated for presence and location fas‐1 gene, which codes an isopentenyl transferase, committed step in cytokinin biosynthesis. The was determined by PCR using a set primers to most conserved regions gene Southern hybridization genomic digests fragment as probe. Both methods revealed 18 virulent absence avirulent isolates. Thus, there is strong relationship between virulence organism. probing blots linear...
