Sphingosine-1-phosphate (S1P) plays multiple roles in bone metabolism and regeneration. Here, we have identified a novel S1P-regulated osteoanabolic mechanism functionally connecting osteoblasts (OBs) to the highly specialized vasculature. We demonstrate that S1P/S1PR3 signaling OBs stimulates vascular endothelial growth factor (VEGFa) expression secretion promote an autocrine boost osteogenic H-type differentiation of marrow cells paracrine manner. VEGFa-neutralizing antibodies VEGF...

Red blood cells (RBC) are the major carriers of sphingosine-1-phosphate (S1P) in blood. Here we show that variations RBC S1P content achieved by altering synthesis and transport genetic pharmacological means regulate glucose uptake metabolic flux. This is due to S1P-mediated activation catalytic protein phosphatase 2 (PP2A) subunit leading reduction cell-surface transporters (GLUTs). The mechanism dynamically responds cues from environment increasing synthesis, enhancing PP2A activity,...

Abstract Membrane phospholipids affect GLUT function in RBC but the role of their large intercellular S1P pool is unknown. We show that altering content by intervening its precursor uptake, synthesis and efflux had major impacts on glucose uptake glycolysis. identified mechanism as direct activation PP2A leading to cell-surface reduction. This was highly dynamic it responded metabolic cues from environment regulating intracellular thereby hence counteracting pathophysiological conditions...
