Native chemical ligation followed by desulfurization is a powerful strategy for the assembly of proteins. Here we describe development high-yielding, one-pot ligation−desulfurization protocol that uses trifluoroethanethiol (TFET) as novel thiol additive. The synthetic utility this TFET-enabled methodology demonstrated efficient multi-step syntheses two tick-derived proteins, chimadanin and madanin-1, without need any intermediary purification.
The transport of anions across cellular membranes is an important biological function governed by specialised proteins. In recent years, many small molecules have emerged that mimick the anion behaviour these proteins, but only a few synthetic also display gating/switching seen in systems. A series thiosquaramides was synthesised and their pH-dependent chloride binding investigated using 1H NMR titrations, single crystal X-ray diffraction variety vesicle-based techniques. Spectrophotometric...
Asp-ecially useful: A synthetic β-mercapto aspartate residue facilitates the rapid ligation to a range of peptide thioesters. Following reaction (and without purification), chemoselective desulfurization moiety in presence unprotected cysteine residues afforded native products.
A number of macrocyclic squaramide-containing receptors (MSQs) have been designed and synthesised their interaction with a range inorganic anions was studied in solution by 1H NMR spectroscopy ESI-HRMS. The binding data revealed remarkable sulfate aqueous mixtures from 0.5 to 50% v/v H2O/DMSO-d6. larger [3]-MSQs were found better match the size shape ion than [2]-MSQs, providing high affinity selectivity for while other tetrahedral divalent such as selenate, phosphate species chromate...
The transmembrane transport of anions by small synthetic molecules is a growing field in supramolecular chemistry and has focussed mainly on the chloride. On other hand, highly hydrophilic sulfate anion across lipid bilayers much less developed, even though inability to cellular membranes been linked variety genetic diseases. Tris-thioureas possess high affinities have shown be excellent chloride bicarbonate transporters. Herein we report abilities series tris-ureas tris-thioureas based...
Functional drug carrier systems have potential for increasing solubility and potency of drugs while reducing side effects. Complex polymeric materials, particularly anisotropic structures, are especially attractive due to their long circulation times. Here, we conjugated cyclic peptides the biocompatible polymer poly(2-hydroxypropyl methacrylamide) (pHPMA). The resulting conjugates were functionalized with organoiridium anticancer complexes. Small angle neutron scattering static light...
A cyclic peptide based receptor, bearing two dipicolylamino arms complexed to zinc(II) ions, binds pyrophosphate ions with high affinity and selectivity in aqueous solution as determined using an indicator displacement assay.
Into the void: A large, cavity-containing trinuclear triangle with accessible binding sites has been synthesized, structurally characterized, and incorporated into a chiral metal–organic framework (see picture). The resulting 3D framework, trifunctional amine bridging group, void volume of 56 % that is to solvent adsorbs variety gases various degrees. Detailed facts importance specialist readers are published as "Supporting Information". Such documents peer-reviewed, but not copy-edited or...
A library of anion receptors comprising cyclic peptide scaffolds bearing two (ZnII-DPA) binding sites have been synthesised and their abilities evaluated using colourimetric indicator displacement assays with three different indicators. The resulting chemosensing ensembles provided excellent discrimination between polyphosphate ions, several the receptor : combinations providing naked-eye sensing pyrophosphate ions in aqueous solutions containing more than 100 fold excess ATP.
The partition and self-assembly of a new generation cyclic peptide-polymer conjugates into well-defined phospholipid trans-bilayer channels is presented. By varying the structural parameters through ligation hydrophobic hydrophilic polymers, both structure artificial using large unilamellar vesicle assays required for bilayer partitioning are elucidated. In addition, temperature was used as an external stimulus modulation transbilayer channel formation without requiring redesign synthesis...
We review the advantages and drawbacks of various synthetic strategies for conjugating peptides to polymers obtained from reversible-deactivation radical polymerization (also known as living polymerization, controlled or controlled/living polymerization). By using a selection examples, we summarize concept behind divergent convergent syntheses, which lead production linear grafted peptide/polymer conjugates. In second section this review, present an overview near–universal approach...
We have established a new strategy to produce functional organic nanotubes of controlled structure from cyclic peptide/polymer conjugates. The guiding peptide motif was coupled the polymeric chains viacopper-catalyzed azide–alkyne click reaction, yield very well-defined resulting conjugates were then self-assembled into in solution. able control some extent length nanotubes, by modifying polymer chain as well mixing different molecular weights together. In similar fashion we prepare tubes...
ConspectusPhosphate oxoanions and phosphorylated biomolecules (such as nucleotides, lipids, proteins) play key roles in a wide range of biological processes. The ability to selectively detect these ions the presence each other has numerous applications biochemistry biomedicine. However, receptors sensors that can discriminate between polyphosphate species with high selectivity biologically relevant conditions are rare.In this Account, we show how peptides (both cyclic linear) be used...
We report a rotaxane based on simple urea motif that binds Cl- selectively as separated ion pair with H+ and reports the anion binding event through fluorescence switch-on response. The host over more basic anions, which deprotonate framework, less bind weakly. mechanical bond also imparts size selectivity to ditopic host.
A review of fluorescent sensing arrays for anions and cations, highlighting promising strategies directions future research.
Lipophilic macrocycles efficiently extract sulfate ions from water into chloroform and transport this ion across a bulk liquid membrane in the presence of competing anions (chloride, nitrate dihydrogenphosphate).
A total of 72 hydrogen bonds are formed in the spontaneous association calix[4]arene tetramelamine and barbituric acid derivatives to give nanosized assemblies type represented picture. These consist 15 components that assemble a completely diastereoselective sense: eight possible diastereomers only all-staggered diastereoisomer is obtained.
