Ghrelin, the endogenous ligand for GH secretagogue receptors, has been reported to influence acid gastric secretion and motility, but its potential gastroprotective effect is unknown. The aims of this study were 1) examine effects central peripheral administration ghrelin on ethanol-induced ulcers in conscious rats, 2) investigate possible roles nitric oxide (NO), vagal nerve, sensory fibers ghrelin. Ghrelin was administered either intracerebroventricularly or sc 30 min before ethanol,...
It has previously been reported that growth hormone secretagogues (GHS) may have a role in the regulation of bone metabolism animals and humans. In this study we evaluated effect ghrelin, endogenous ligand GHS receptors, on proliferation rate osteoblast activity primary cultures rat calvaria osteoblasts. same experiments, compared effects ghrelin with those hexarelin (HEXA) EP-40737, two synthetic different characteristics. Both HEXA (10 −11 −10 −8 M) significantly stimulated at low...
We examined the possible central and peripheral effects of synthetic growth hormone secretagogues (GHS), hexarelin (Hexa) EP 40737 (D-Thr-D-Trp (2-Me)-Ala- Trp-D-Phe-Lys-NH2), their endogenous counterpart, ghrelin, on gastric acid secretion. The compounds were administered intracerebroventricularly (i.c.v.) or subcutaneously (s.c.) in conscious male rats volume secretion output 3 h after pylorus ligation (Shay-test). Central Hexa, ghrelin administration (from 0.1 pmol to 1 nmol/rat, i.c.v.)...
Background and purpose: Ghrelin, a gut‐brain peptide, is considered gastroprotective factor in gastric mucosa. We investigated the role of prostaglandins (PG) possible interplay between PGs nitric oxide (NO) ghrelin gastroprotection against ethanol (EtOH)‐induced lesions. Experimental approach: examined effects (1) central (4 μg per rat) injection on PGE 2 accumulation normal or EtOH–lesioned mucosa, (2) pretreatment with indomethacin (10 mg kg −1 , p.o.), non‐selective cyclooxygenase (COX)...
The effect of amylin on gastric ulcers induced by oral administration indomethacin (Indo, 20 mg kg −1 at a dosing volume 5 ml) or ethanol 50% (EtOH, 1 ml/rat) was investigated in conscious rats. Amylin given intracerebroventricularly (0.22, 0.66 and 2.2 μg/rat, i.c.v.) demonstrated dose‐dependent cytoprotective against both Indo EtOH‐induced ulcers. In contrast, amylin, subcutaneously doses effective inhibiting acid secretion (2.5, 10 40 μg , s.c), did not show any effect. interaction...
Natural antioxidants are considered as promising compounds in the prevention/treatment of osteoporosis. We studied ability purified δ-tocotrienol (δ-TT) isolated from a commercial palm oil (Elaeis guineensis) fraction to protect osteoblast MC3T3-E1 and osteocyte MLO-Y4 cells against tert-butyl hydroperoxide (t-BHP)-induced oxidative damage mechanisms involved its protective action MC3T3-E1.MC3T3-E1 were treated with δ-TT (1.25-20 µg/ml for 2 h) followed by t-BHP at 250 µM or 125 3 h,...
Vgf gene expression has been detected in various endocrine and neuronal cells the gastrointestinal tract. In this study we investigated pharmacological activity of different VGF-derived peptides. Among these, TLQP-21, corresponding to 556-576 fragment protein was unique active peptide, its profile further studied. The effects TLQP-21 were examined vitro by smooth muscle contraction isolated preparations from rat tract and, vivo, assessing gastric emptying rats. Rat stomach tissues also...
Recent research suggests a role for ghrelin in the modulation of inflammatory disorders. However, type receptor (GHS-R) involved both anti-inflammatory and anti-hyperalgesic actions remains to be characterized. In this study, we examined whether inhibitory effect development hyperalgesia edema induced by intraplantar carrageenan administration depends on an interaction with GHS-R1a. Both central (1 nmol/rat, i.c.v.) peripheral (40 nmol/kg, i.p.) selective GHS-R1a agonist EP1572 had no...
Abstract The ‘fate’ of growth plate hypertrophic chondrocytes has been long debated with two opposing theories: cell apoptosis or survival transformation into osteogenic cells. This study was carried out on the proximal tibial rabbits using light microscopy, scanning and transmission electron microscopy. We focused particularly orientation specimens included in order to define mineral deposition vascular invasion lines obtain histological ultrastructural images at corresponding height plate....
Cimetidine, an H2 receptor antagonist, administered into a lateral ventricle of the rat brain caused significant increase in tail flick latency. Dimaprit, specific agonist, failed to counteract analgesic effect cimetidine. In contrast, it enhanced cimetidine and per se had marked activity. The opioid antagonist naloxone was without effect. Pretreatment with CaCl2 completely prevented action These findings suggest that dimaprit is not due effects on receptors.
An estrogen-regulated cytoplasmic protein has been purified from MCF-7 human breast cancer cells with anion exchange and monoclonal antibody affinity chromatographies. The hasa monomeric mol wt of 28,000 isoelectric species pi's between 5.9 6.0. Amino acid analysis indicates the is acidic, probably hydrophilic, contains unusually low amounts methionine half cystine. This rapid, two-step procedure produces in high yield demonstrates power antibodies purification.
Ghrelin, the endogenous ligand of GH secretagogue receptor (GHS-R) has been previously shown to inhibit gastric acid secretion in pylorus-ligated rats. Two isoforms GHS-R have identified: GHS-R(1a) and GHS-R(1b). The present study aimed: (i) characterise type involved central inhibitory activity ghrelin by using des-octanoyl ghrelin, synthetic agonist (EP1572) antagonist (D-Lys(3)-GHRP-6) (ii) investigate relationship between cortistatin (CST) control natural neuropeptide CST-14 octapeptide...
Activated neutrophils (PMNs), the ROS/RNS released by PMNs and derived inflammatory processes are involved in pathogenesis progression of human airway diseases. Similar diseases also present horses which suffer from recurrent obstruction (RAO), exercise-induced pulmonary haemorrhage (EIPH) (IAD). Hyaluronic acid (HA) plays numerous roles modulating processes. The aim this study was to examine whether a preparation HA (MW 900 000 Da) interferes with during course equine PMN respiratory...
Bone remodeling and repair require osteogenic cells to reach the sites that need be rebuilt, indicating stimulation of osteoblast migration could a promising osteoanabolic strategy. We showed purified δ-tocotrienol (δ-TT, 10 μg/mL), isolated from commercial palm oil (Elaeis guineensis) fraction, stimulates both MC3T3-E1 osteoblast-like primary human bone marrow mesenchymal stem (BMSC) as detected by wound healing assay or Boyden chamber respectively. The ability δ-TT promote is dependent on...
