Mitofusin 2 (Mfn2), mitochondrial outer membrane protein which is involved in rearrangement of these organelles, was first described pathology hypertension and diabetes, more recently much attention paid to its functions Charcot-Marie-Tooth type 2A neuropathy (CMT2A). Here, cellular energy metabolism investigated mouse embryonic fibroblasts (MEF) differing the presence Mfn2 gene; control (MEFwt) with gene depleted MEFMfn2-/-. These two cell lines were compared terms various parameters...
Abstract Duchenne muscular dystrophy (DMD) is characterized by progressive muscle degeneration and neuropsychiatric abnormalities. Loss of full-length dystrophins both necessary sufficient to initiate DMD. These isoforms are expressed in the hippocampus, cerebral cortex (Dp427c), cerebellar Purkinje cells (Dp427p). However, our understanding consequences their absence, which crucial for developing targeted interventions, remains inadequate. We combined RNA sequencing with genome-scale...
Abstract In attempts to develop effective therapeutic strategies limit post-ischemic injury, mitochondria emerge as a key element determining neuronal fate. Mitochondrial damage can be alleviated by various mechanisms including mitochondrial network remodelling, elimination and protein biogenesis. However, the regulating relationships between these phenomena are poorly understood. We hypothesized that mitofusin 2 (Mfn2), GTPase involved in fusion, trafficking endoplasmic reticulum (ER)...
Charcot-Marie-Tooth disease type 2A (CMT2A) is an autosomal dominant axonal peripheral neuropathy caused by mutations in the mitofusin 2 gene (MFN2). Mitofusin a GTPase protein present outer mitochondrial membrane and responsible for regulation of network architecture via fusion mitochondria. As that process known to be strongly dependent on activity 2, it postulated MFN2 mutation within domain may lead impaired activity, turn dysfunction. The work described here has therefore sought verify...
Transient ischemic attacks (TIA) result from a temporary blockage in blood circulation the brain. As TIAs cause disabilities and often precede full-scale strokes, effects of TIA are investigated to develop neuroprotective therapies. We analyzed changes mitochondrial network dynamics, mitophagy biogenesis sections gerbil hippocampus characterized by different neuronal survival rate after 5-minute ischemia-reperfusion (I/R) insult. Our research revealed significantly greater mtDNA/nDNA ratio...
Mitochondria are multifunctional organelles that participate in numerous processes response to viral infection, but they also a target for viruses. The aim of this study was define subcellular events leading alterations mitochondrial morphology and function during infection with ectromelia virus (ECTV). We used two different cell lines combination immunofluorescence techniques, confocal electron microscopy, flow cytometry address changes following infection. Early L929 fibroblasts RAW 264.7...
Several studies have provided evidence that complex relationships between autophagic and apoptotic cell death pathways occur in cancer virus-infected cells. Previously, we demonstrated infection of macrophages with Moscow strain ectromelia virus (ECTV-MOS) induces apoptosis under vitro vivo conditions. Here, found autophagy was induced RAW 264.7 cells during ECTV-MOS. Silencing beclin 1, an autophagy-related gene, reduced the percentage late macrophages. Pharmacological modulation by...
Ectromelia virus (ECTV, the causative agent of mousepox), which represents same genus as variola (VARV, responsible for smallpox in humans), has served years a model studying mechanisms poxvirus-induced disease. Despite increasing knowledge on interaction between ECTV and its natural host-the mouse-surprisingly, still little is known about cell biology infection. Because pathogen with cytoskeleton growing area research virus-host interplay, aim present study was to evaluate consequences...
Transient brain ischemia in gerbils is a common model to study the mechanisms of neuronal changes hippocampus. In cornu ammonnis 2–3, dentate gyrus (CA2-3,DG) regions hippocampus, neurons are resistant 5-min ischemia/reperfusion (I/R) insult, while 1 (CA1) found be I/R-vulnerable. The quantitative polymerase chain reaction (qRT-PCR) widely used expression genes involved these phenomena. It requires stable and reliable for normalization, which crucial comparable reproducible analyses...
Abstract Autophagy is a self-degradation process of cellular components. It plays both antiviral and pro-viral roles in the life cycle different viruses pathogenesis viral diseases. In this study, we evaluated autophagy induction splenocytes ectromelia virus (ECTV)-resistant C57BL/6 ECTV-susceptible BALB/c mice during infection with Moscow strain (ECTV-MOS). was analyzed using Western blot method by assessing type II microtubule-associated protein 1 (MAP1) light chain 3 (LC3) Beclin...
The purpose of the study was to evaluate synthesis IFN-γ, IL-2, TNF-α (Th1/Tc1) and IL-4 (Th2/Tc2) at CD4+ T CD8+ cell level in BALB/c C57BL/6 mice course infection with ectromelia virus Moscow strain (ECTV-MOS).Synthesis cells draining lymph nodes (DLNs) spleens detected by intracellular staining flow cytometry analysis. Our results showed an increase percentage IFN-γ -synthesizing only DLNs early stages infection. Moreover, IL-2 occurred earlier stronger compared mice. mainly infected...
During mousepox in resistant (C57BL/6) or susceptible (BALB/c) strains of mice, stimulation Th1 Th2 cytokine immune response, respectively, is observed. Because mechanisms different polarization T cells remain elusive, this study, we quantitatively assessed the phenotype antigen-presenting (APCs) involved ectromelia virus (ECTV) antigen presentation and cluster formation with effector secondary lymphoid organs BALB/c C57BL/6 mice. We showed that both mice display similar dynamics kinetics...
Induction of autophagy by ectromelia virus (ECTV) in primary cultures bone marrow-derived macrophages (BMDMs) was investigated. The results showed that ECTV infection BMDMs resulted increased formation autophagosomes, level LC3-II protein present aggregates and extensive cytoplasmic vacuolization. These data indicate an autophagic activity during infection.
