The Notch signaling pathway plays a crucial role in skeletal muscle regeneration mammals by controlling the transition of satellite cells from quiescence to an activated state, their proliferation, and commitment toward myotubes or self-renewal. O-fucosylation on receptor epidermal growth factor (EGF)-like repeats is catalyzed protein O-fucosyltransferase 1 (Pofut1) primarily controls interaction with its ligands. To approach myogenesis, we analyzed murine myoblastic C2C12 cell line...

The chromosome translocations generating PAX3-FOXO1 and PAX7-FOXO1 chimeric proteins are the primary hallmarks of paediatric fusion-positive alveolar subtype Rhabdomyosarcoma (FP-RMS). Despite ability these transcription factors to remodel chromatin landscapes promote expression tumour driver genes, they only inefficiently malignant transformation in vivo . reason for this is unclear. To address this, we developed an ovo model follow response spinal cord progenitors PAX-FOXO1s. Our data...

Urban pollution is known to have major adverse effects on lung health. To assess the effect of alveolar regeneration, we exposed adult mice a simulated high-pollution urban atmosphere, using an innovative CESAM simulation chamber (Multiphase Atmospheric Experimental Simulation Chamber, https://cesam.cnrs.fr/ ). We demonstrated that atmosphere alters mesenchymal stem cell niche properties by inducing lipofibroblast myofibroblast shift and induces epithelial dysfunction.

Obesity is associated with oxidative stress, a major factor in carcinogenesis, and high leptin concentration. The aim of this study was to determine the effects on antioxidant response three human mammary epithelial cells each presenting different neoplastic status: healthy (HMEC), oestrogen-receptor positive MCF-7 triple-negative MDA-MB-231 cells. This vitro kinetic characterized cell after 1, 6 24 h presence (10 or 100 ng/ml).The defined terms glutathione content, gene expression catalytic...

The protein O-glucosyltransferase 1 (Poglut1) links O-glucose to epidermal growth factor-like repeats harboring the C1XSX(P/A)C2 consensus sequence. Poglut1 is a ubiquitous endoplasmic reticulum-resident largely found in metazoans, but only about 50 proteins possess this Among them, Notch receptors have multiple O-glucosylation sites and their activation depends on status. In adult skeletal muscle, signaling contributes maintenance of satellite cell (SC) quiescence proliferation myoblasts...

Past work has shown that the protein O-fucosyltransferase 1 (POFUT1) is involved in mammal myogenic differentiation program. Pofut1 knockdown (Po –) murine C2C12 cells leads to numerous elongated and thin myotubes, suggesting significant defects secondary fusion. Among few pathways this process, NFATc2/IL-4 described as major one. To unravel impact of POFUT1 on fusion, we used wild-type (WT) Po – cell lines follow Myf6, Nfatc2, Il-4 Il-4rα expressions during a 120 h time course. Secreted...

SUMMARY Muscle stem cells (MuSCs) are essential for skeletal muscle repair. Following injury, MuSCs reside in low oxygen environments until fibers and vascularization restablished. The dynamics of levels during the regenerative process its impact on repair has been underappreciated. We confirm that is initiated a environment followed by gradual reoxygenation. Strikingly, when reoxygenation limited keeping mice under systemic hypoxia, impaired leads to formation hypotrophic myofibers. In vivo...

Introduction: Air pollution (AP) represents the largest environmental risk for health. Such considerations however rely on static quantification of only a few individual components AP, without considering their interactions, chemical reactivity and synergistic toxicity. We therefore developed an innovative approach to analyze health effects "real life" model atmospheres at preclinical level. Here, we report first results feasibility study. Methods: A realistic atmosphere,...
