Immunoglobulin class switch recombination (CSR) plays critical roles in controlling infections and inflammatory tissue injuries. Here, we show that AFF3, a candidate gene for both rheumatoid arthritis type 1 diabetes, is molecular facilitator of CSR with an isotype preference. Aff3-deficient mice exhibit low serum levels immunoglobulins, predominantly immunoglobulin G2c (IgG2c) followed by IgG1 IgG3 but not IgM. Furthermore, weak resistance to Plasmodium yoelii infection, confirming Aff3...

Summary Lipoprotein lipase ( LPL ) mRNA expression in chronic lymphocytic leukaemia (CLL) is associated with an unmutated immunoglobulin profile and poor clinical outcome. We evaluated the subcellular localization of protein CLL cells that did or not express mRNA. Our results show differently located depending on whether it incorporated from extracellular medium mutated generated de novo by leukaemic patients. The specific quantification endogenous correlates levels mutational IGHV status,...
Introduction BAP1 is a deubiquitinase (DUB) of the Ubiquitin C-terminal Hydrolase (UCH) family that regulates gene expression and other cellular processes, through its direct catalytic activity on repressive epigenetic mark histone H2AK119ub, as well several substrates. also highly important tumor suppressor, expressed functional across many cell types tissues. In recent work, we demonstrated intrinsic role in B lineage development murine bone marrow, however regulation mediated humoral...
Abstract Activation induced deaminase (AID) initiates class switch recombination (CSR) and somatic hypermutation of the antibody genes but also mutates “off-target” genes, with oncogenic consequences. Understanding mechanisms that render such loci susceptible to AID is therefore necessary. We identify H3K79 histone methyltransferase DOT1L as a nuclear AID-proximal factor required for CSR off-target activity, including IgH-cMyc translocations. Mechanistically, limits promoter-proximal...
