FhaC is an integral outer membrane protein of the whooping cough agent Bordetella pertussis that mediates transport to cell surface a major virulence factor, filamentous haemagglutinin adhesin FHA. The FHA/FhaC pair prototypic TpsA/TpsB system widespread 'Two-Partner Secretion' pathway, dedicated long extracellular proteins in various pathogenic and environmental Gram-negative bacteria. belongs ubiquitous Omp85 superfamily transporters. X-ray structure shows transmembrane β-barrel channel...
Bacterial contact-dependent growth inhibition (CDI) systems use a type Vb secretion mechanism to export large CdiA toxins across the outer membrane by dedicated transporters called CdiB. Here, we report first crystal structures of two CdiB from Acinetobacter baumannii and Escherichia coli . adopt TpsB fold, containing 16-stranded transmembrane β-barrel connected periplasmic domains. The lumen pore is occluded an N-terminal α-helix conserved extracellular loop 6; these elements different...
Omp85 transporters mediate protein insertion into, or translocation across, membranes. They have a conserved architecture, with POTRA domains that interact substrate proteins, 16-stranded transmembrane β barrel, and an extracellular loop, L6, folded back in the barrel pore. Here using electrophysiology, vivo biochemical approaches electron paramagnetic resonance, we show L6 loop of transporter FhaC changes conformation modulates channel opening. Those conformational involve breaking...
In this study, we develop a branch and bound solution algorithm to solve the workload smoothing problem. Our incorporates new formulas for dynamically computing lower on optimal value of objective function determining earliest workstations tasks. It also uses fast heuristic good initial upper bound. A comprehensive experimental analysis is conducted in study. The demonstrates outstanding performance its efficiency solving medium-sized problems.
The Two-Partner secretion pathway mediates protein transport across the outer membrane of Gram-negative bacteria. TpsB transporters belong to Omp85 superfamily, whose members catalyze insertion into, or translocation membranes without external energy sources. They are composed a transmembrane β barrel preceded by two periplasmic POTRA domains that bind incoming substrate. Here we used an integrative approach combining in vivo assays, mass spectrometry, nuclear magnetic resonance and electron...
Workload smoothing techniques have been used to establish a balance among stations in assembly lines by assigning the tasks evenly stations. Smoothing facilitates planning workforce and optimizing resource allocation problems. Considering these advantages, we address simple line this research develop problem specific heuristic algorithm solve large instances effectively. We test effectiveness of with experimental study.
