The mRNA translation initiation region (TIR) comprises the initiator codon, Shine-Dalgarno (SD) sequence and translational enhancers. Probably most abundant class of enhancers contains A/U-rich sequences. We have tested influence SD length presence on efficiency initiation. found that during bacterial growth at 37°C, a six-nucleotide (AGGAGG) is more efficient than shorter or longer enhancer contributes strongly to initiation, having greatest stimulatory effect in exponential phase bacteria....

Resistance proteins are perceived as mechanisms protecting bacteria from the inhibitory effect of their produced antibiotics or competitors. Here, we report that antibiotic resistance regulate lincomycin biosynthesis in response to subinhibitory concentrations antibiotics.

Detailed mutational analysis examines the roles of individual residues Vga(A) linker in determining antibiotic resistance phenotype. It defines a narrowed region 212 to 220 whose composition determines specificity lincosamides, pleuromutilins, and/or streptogramins A. From analogy with recently described function homologous ABC-F protein EttA as translational factor, we infer that interacts ribosome and directly or indirectly affects binding respective antibiotic.

Vga(A) protein variants confer different levels of resistance to lincosamides, streptogramin A, and pleuromutilins (LSAP) by displacing antibiotics from the ribosome. Here, we show that expression vga(A) Staphylococcus haemolyticus is regulated cis-regulatory RNA in response LSAP mechanism ribosome-mediated attenuation. The specificity induction depends on Vga(A)-mediated rather than sequence riboregulator. Fine tuning between activity its may contribute selection more potent because newly...

vanZ, a member of the VanA glycopeptide resistance gene cluster, confers to lipoglycopeptide antibiotics independent cell wall precursor modification by vanHAX genes. Orthologs vanZ are present in genomes many clinically relevant bacteria, including Enterococcus faecium and Streptococcus pneumoniae; however, genes absent Staphylococcus aureus. Here, we show that expression enterococcal paralogs S. aureus increases minimal inhibitory concentrations teicoplanin, dalbavancin, oritavancin new...

The macrolide antibiotic erythromycin binds at the entrance of nascent peptide exit tunnel large ribosomal subunit and blocks synthesis peptides longer than between six eight amino acids. Expression a short open reading frame in 23 S rRNA encoding five acids confers resistance to by mechanism that depends strongly on both sequence length peptide. In this work we have used cell-free system for protein with components high purity clarify molecular basis mechanism. We found interacts...

Summary Expression of specific short peptides can render cells resistant to macrolide antibiotics. Peptides conferring resistance structurally different macrolides including oleandomycin, azithromycin, azaerythromycin, josamycin and a ketolide cethromycin were selected from random pentapeptide expression library. Analysis the entire collection allowed their classification into five distinct groups according sequence similarity type they confer. A strong correlation was observed between...

A new, efficient, and general semisynthesis of hydnocarpin-type flavonolignans was developed optimized, enabling gram-scale production hydnocarpin D (2). Moreover, the syntheses optically pure isomers [(10R,11R)-hydnocarpin (1a), (10R,11R)-hydnocarpin (2a), (10S,11S)-hydnocarpin (2b)], as well synthesis isohydnocarpin (8), were achieved for first time utilizing this new method. Abstract Patients infected with SARS-CoV-2 risk co-infection Gram-positive bacteria, which severely affects their prognosis. Antimicrobial drugs dual antiviral and antibacterial activity would be very useful in this setting. Although glycopeptide antibiotics are well-known as strong drugs, some of them also active against RNA viruses like SARS-CoV-2. It has been shown that the efficacy can enhanced by synthetic modifications. We here report synthesis biological evaluation seven derivatives...

Abstract In natural environments, antibiotics are an important instrument of inter-species competition. At subinhibitory concentrations, they act as cues or signals inducing antibiotic production: however, our knowledge well-documented antibiotic-based sensing systems is limited.
