New polyanionic modifications of polyamide nucleic acid mimics were obtained. Thymine decamers synthesized from respective chiral α- and γ-monomers, their enantiomeric purity was assessed. Here, we present the decamer synthesis, purification characterization by MALDI-TOF mass spectrometry an investigation hybridization properties decamers. We show that modified γ-S-carboxyethyl-T10 PNA forms a stable triplex with polyadenine DNA.
Abstract Regioselective method for 1-β-D-glucopyranosylthymine incorporation into oligonucleotides has been developed and substrate properties of the latters in DNA synthesis hydrolysis reactions were investigated.
Inorganic polyphosphates (polyP), according to literature data, are involved in the regulatory processes of molecular complex Saccharomyces cerevisiae cell wall (CW). The aim work was reveal relationship between polyP, acid phosphatase Pho3p, and major CW protein, glucanosyl transglycosylase Bgl2p, which is main glucan-remodelling enzyme with amyloid properties. It has been shown that yeast cells deletion PHO3 gene contain more high alkali-soluble polyP also resistant exposure alkali...
Abstract A method of completely chemical synthesis 3′-azido-3′-deoxythymidine-terminated oligonucleotides via 5′-H-phosphonate AZT is described.
The study is devoted to 8-oxo-2'-deoxyguanosine (8-oxo-dG), which used as a biomarker of oxidative damage DNA and its associated diseases. based on the hypothesis put forward by authors about ability 8-oxo-dG, main product oxidation, act signal molecule that activates cell resistance nonspecific stress repair. In addition studying molecular mechanisms action have carried out an investigation effects they caused organism level.
Abstract An oligodeoxynucleotide bearing a 3′-terminal perylene-containing pseudonucleoside unit was synthesized and used as probe in homogeneous hybridization. The fluorescence anisotropy of the perylene dye rose upon hybridization modified conjugate with complementary nucleotide sequence. These results provide for designing efficacious probes.
The changes of the 8-oxo-2'-deoxyguanosine (8-oxo-dG)/dG ratio in DNA laboratory mice hepatocytes under influence toxic stress were studied. It was shown that injection carbon tetrachloride causes growth 8-oxo-dG level. A rapid increase level occurs during first day experiment at short-term stress. Subsequently, 48 hours after initial injection, decreases to control values. This change value biomarker can be attributed activation reparative and antioxidant systems. subsequent results again...
