Abstract In this study the exchange between 1 H magnetization in “free” water ( f ) and that a pool with restricted motion r was observed tissues vivo using NMR saturation transfer methods. Exchange these two pools demonstrated by decrease steady‐state relaxation times of radiofrequency irradiation . A method is described for the correction of geometric distortions occurring in echo planar images. The are caused large part by static magnetic field inhomogeneities, leading to pixel shifts, particularly phase encode direction. By characterizing inhomogeneities from a map, image can be unwarped so that accurate alignment conventionally collected images made. algorithm perform unwarping described, and results at 1.5 4 Tesla shown.
Metabolic rate and the subsequent production of reactive oxygen species are thought to contribute aging in a wide range species. The target rapamycin (TOR) is well conserved serine/threonine kinase that regulates cell growth response nutrient status. Here we demonstrate mammalian cells TOR (mTOR) pathway plays significant role determining both resting consumption oxidative capacity. In particular, level complex formation between mTOR one its known protein partners, raptor, correlated with...
Ca(2+) has been postulated as a cytosolic second messenger in the regulation of cardiac oxidative phosphorylation. This hypothesis draws support from well-known effects on muscle activity, which is stimulated parallel with Ca(2+)-sensitive dehydrogenases (CaDH). The phosphorylation were further investigated isolated porcine heart mitochondria at level metabolic driving force (NADH or Deltapsi) and ATP production rates (flow). resulting force-flow (F-F) relationships permitted analysis...
The mechanism that concentrates the urine to an osmolality several times of systemic plasma results in high concentrations solutes (particularly NaCl and urea) extracellular fluid renal medulla, but not labyrinth cortex.Intracellular must be equal animals, known intracellular levels Na K salts urea medullas are much too low balance osmolality. The purpose these studies was identify other osmolytes present.Cortexes from rabbit rat kidneys were analyzed by proton nuclear magnetic resonance,...
Background Commercial low-field-strength MRI systems are generally not equipped with state-of-the-art hardware, and suitable for demanding imaging techniques. An system was developed that combines low field strength (0.55 T) high-performance technology. Purpose To evaluate applications of a system, specifically MRI-guided cardiovascular catheterizations metallic devices, diagnostic in high-susceptibility regions, efficient image acquisition strategies. Materials Methods A commercial 1.5-T...
Calcium is believed to regulate mitochondrial oxidative phosphorylation, thereby contributing the maintenance of cellular energy homeostasis. Skeletal muscle, with an conversion dynamic range up 100-fold, extreme case for evaluating balance ATP production and consumption. This study examined role Ca(2+) in entire phosphorylation reaction network isolated skeletal muscle mitochondria attempted extrapolate these results back vivo. Kinetic analysis was conducted evaluate dose-response effect on...
Cardiomyocytes are organized in microstructures termed sheetlets that reorientate during left ventricular thickening. Diffusion tensor cardiac magnetic resonance (DT-CMR) may enable noninvasive interrogation of vivo microstructural dynamics. Dilated cardiomyopathy (DCM) is a condition abnormal myocardium with unknown sheetlet function. This study sought to validate DT-CMR measures microstructure against histology, characterize dynamics wall thickening, and apply the technique hypertrophic...
Solution pH was measured using water proton NMR via chemical exchange dependent saturation transfer (CEST) with selected sites. Several useful pH-sensitive agents were found: 5,6-dihydrouracil, 5-hydroxytryptophan, and a combination of 5-hydroxytryptophan 2-imidazolidinethione. A ratiometric approach developed that permitted determinations independent T(1) or site concentration.
Background— Managing chest pain in the emergency department remains a challenge with current diagnostic strategies. We hypothesized that cardiac MRI could accurately identify patients possible or probable acute coronary syndrome. Methods and Results— The performance of was evaluated prospective study 161 consecutive patients. Enrollment required 30 minutes compatible myocardial ischemia but an ECG not infarction. performed at rest within 12 hours presentation included perfusion, left...
In nine subjects with carotid atherosclerosis, double-oblique, contrast material–enhanced, double inversion-recovery, fast spin-echo magnetic resonance (MR) images were acquired through atheroma in the proximal internal artery. Fibrocellular tissue within selectively enhanced 29% after administration of gadolinium-based agent. Contrast enhancement helped discriminate fibrous cap from lipid core a contrast-to-noise ratio as good or better than that T2-weighted MR but approximately twice...
PURPOSE: To compare fluorescent microsphere measurements of myocardial blood flow (MBF) with qualitative, semiquantitative, and fully quantitative first-pass perfusion at magnetic resonance (MR) imaging. MATERIALS AND METHODS: Coronary artery occlusion or intracoronary adenosine infusion was successfully performed in 16 beagles; both procedures were simultaneously one animal. MBF assessed analysis. First-pass MR imaging during a dual-bolus administration gadopentetate dimeglumine (0.0025...
Post-translational modification of mitochondrial proteins by phosphorylation or dephosphorylation plays an essential role in numerous cell signaling pathways involved regulating energy metabolism and mitochondrion-induced apoptosis. Here we present a phosphoproteomic screen the matrix begin to establish protein phosphorylations acutely associated with calcium ions (Ca2+) porcine heart mitochondria. Forty-five phosphorylated were detected gel electrophoresis−mass spectrometry Pro-Q Diamond...
