For mass production of lactic acid, we newly constructed a transgenic wine yeast strain that included six copies the bovine L-lactate dehydrogenase gene on genome. On fermentation in inexpensive cane juice-based medium, this recombinant reached 122 g/liter and optical purity was 99.9% or higher.
ABSTRACT We developed a metabolically engineered yeast which produces lactic acid efficiently. In this recombinant strain, the coding region for pyruvate decarboxylase 1 ( PDC1 ) on chromosome XII is substituted that of l -lactate dehydrogenase gene LDH through homologous recombination. The expression mRNA genome-integrated regulated under control native promoter, while completely disrupted. Using method, we constructed diploid transformant, with each haploid genome having single insertion...
(E, E, E)-Geranylgeraniol (GGOH) is a valuable starting material for perfumes and pharmaceutical products. In the yeast Saccharomyces cerevisiae, GGOH synthesized from end products of mevalonate pathway through sequential reactions farnesyl diphosphate synthetase (encoded by ERG20 gene), geranylgeranyl synthase (the BTS1 some endogenous phosphatases. We demonstrated that overexpression diacylglycerol phosphatase (DPP1) gene could promote production. also found BTS1-DPP1 fusion was more...
A plant- and crop-based renewable plastic, poly-lactic acid (PLA), is receiving attention as a new material for sustainable society in place of petroleum-based plastics. We constructed metabolically engineered Saccharomyces cerevisiae that has both pyruvate decarboxylase genes (PDC1 PDC5) disrupted the genetic background to express two copies bovine L-lactate dehydrogenase (LDH) gene. With this recombinant, yield lactate was 82.3 g/liter, up 81.5% glucose being transformed into lactic on...
Office environments play a critical role in employee wellbeing and productivity. While the benefits of incorporating nature into workspaces have been recognized, specific visual characteristics that contribute to restorativeness remain unclear. This study investigates how office environments, specifically presence greenery color complexity, are associated with perceived restorativeness. In Study 1, we developed scale based on Attention Restoration Theory measure restorative consisting three...
The yeast Saccharomyces cerevisiae, a promising host for lignocellulosic bioethanol production, is unable to metabolize xylose. In attempts confer xylose utilization ability in S. number of isomerase (XI) genes have been expressed heterologously this yeast. Although several these XI encoding were functionally the need still exists cerevisiae strain with improved use commercial production bioethanol. currently much effort has devoted achieve objective, one solutions search new gene that would...
Many terrestrial plants produce large quantities of alkanes for use in epicuticular wax and the pollen coat. However, their carbon chains must be long to useful as fuel or a petrochemical feedstock. Here, we focus on Nymphaea odorata, which produces relatively short its anthers. We identified orthologs Arabidopsis alkane biosynthesis genes AtCER1 AtCER3 N. odorata designated them NoCER1A, NoCER3A NoCER3B. Expression analysis NoCER1A NoCER3A/B cer mutants revealed that enzymes cooperated with...
