ABSTRACT
Exosomes are microvesicles secreted by body cells for intercellular communication. The circular RNA circ_0000338 was found to be present in extracellular vesicles and improve the chemoresistance of colorectal cancer (CRC) cells. However, role exosomal 5-fluorouracil (5-FU) resistance CRC is largely unknown. levels circ_0000338, microRNA 217 (miR-217), miR-485-3p were detected using quantitative real-time PCR (qRT-PCR). 50% inhibitory concentration (IC50) values 5-FU, cell proliferation, apoptosis evaluated counting kit 8 (CCK-8), colony formation, flow cytometry, Western blot assays. interaction between miR-217 or confirmed immunoprecipitation (RIP), dual-luciferase reporter, pulldown isolated ultracentrifugation qualified transmission electron microscopy (TEM), Nanosight tracking analysis (NTA), blotting. Xenograft models performed analyze whether circ_0000338-loaded exosomes could increase 5-FU vivo. level elevated 5-FU-resistant tissues cells, knockdown sensitized through enhancing decreasing proliferation vitro. Mechanistically, directly bound miR-485-3p, inhibition reversed effects on CRC. Additionally, incorporated into transmitted 5-FU-sensitive Treatment-sensitive with containing reduced response both vitro Besides that, higher patients exhibiting showed good diagnostic efficiency delivery via enhanced negative regulation indicating a promising therapeutic marker 5-FU-based chemotherapy patients.
