Abstract When exposed to stress conditions, all cells induce mechanisms resulting in an attempt adapt that involve proteins which, once activated, trigger cell responses by modulating specific signaling pathways. In this work, using a combination of pulldown assays and mass spectrometry analyses, we identified the Neurospora crassa SEB-1 transcription factor binds Stress Response Element (STRE) under heat stress. Orthologs have been functionally characterized few filamentous fungi as being...

Glycogen and trehalose are storage carbohydrates their levels in microorganisms vary according to environmental conditions. In Neurospora crassa, alkaline pH stress highly influences glycogen levels, Saccharomyces cerevisiae, the response also involves calcineurin signaling pathway mediated by Crz1 transcription factor. Recently, yeast, genes were identified as targets of including involved metabolism. this work, we present evidence that N. crassa metabolism is modulated calcium stresses.We...

In Trypanosoma cruzi, the etiologic agent of Chagas disease, Rad51 (TcRad51) is a central enzyme for homologous recombination. Here we describe different roles TcRad51 in DNA repair. Epimastigotes T. cruzi overexpressing TcRAD51 presented abundant TcRad51-labeled foci before gamma irradiation treatment, and faster growth recovery when compared to single-knockout epimastigotes RAD51. Overexpression RAD51 also promoted increased resistance against hydrogen peroxide while exhibited sensitivity...

Significance Circadian rhythms enable organisms to anticipate daily environmental cycles and control the timing of numerous biological processes, including metabolism, optimize health survival organisms. Glycogen metabolism is a conserved glucose homeostatic process; however, molecular mechanisms linking circadian clock glycogen remain largely unknown. In this report, we demonstrate that clock-dependent transcriptional regulation synthase, gsn , regulates oscillations GSN protein...

Environmental pH induces a stress response triggering signaling pathway whose components have been identified and characterized in several fungi. Neurospora crassa shares all six of the Aspergillus nidulans pathway, we investigate here their regulation during an alkaline response. We show that N. pal mutant strains, with exception Δpal-9, which is A. palI homolog, exhibit low conidiation are unable to grow at pH. Moreover, they accumulate pigment melanin, most likely via tyrosinase gene by...

The current treatment of leishmaniasis is based on a few drugs that present several drawbacks, such as high toxicity, difficult administration route, and low efficacy. These disadvantages raise the necessity to develop novel antileishmanial compounds allied with comprehensive understanding their mechanisms action.

The Neurospora crassa NIT-2 transcription factor belongs to the GATA family and plays a fundamental role in regulation of nitrogen metabolism. Because acts by accessing DNA inside nucleus, understanding nuclear import process is necessary characterize its function. Thus, present study, transport was investigated using combination biochemical, cellular, biophysical methods. A complemented strain that produced an sfGFP-NIT-2 fusion protein constructed, localization assessments were made under...

DNA topoisomerases are enzymes that modulate topology. Among them, topoisomerase 3α is engaged in genomic maintenance acting replication termination, sister chromatid separation, and dissolution of recombination intermediates. To evaluate the role this enzyme Trypanosoma cruzi , etiologic agent Chagas disease, a knockout parasite (TcTopo3α KO) was generated, growth, as well its response to several damage agents, were evaluated. There no growth alteration caused by TcTopo3α epimastigote...

Abstract When exposed single-stranded DNA accumulates at stalled or collapsed replication forks, the stress response is triggered to prevent genome instability. Leishmania are parasitic eukaryotes where gene expression universally polycistronic and whose plastic genomes facilitate rapid adaptations in stress, with evidence implicating intrinsic as a source. Little known about response. In this study, we reveal global dynamics of L. major promastigotes by performing ChIP-seq on three key...

Land-use conversion changes soil properties and their microbial communities, which, combined with the overuse of antibiotics in human animal health, promotes expansion resistome. In this context, we aimed to profile resistome microbiota soils under different land practices. We collected eight samples from locations countryside São Paulo (Brazil), assessed community profiles based on 16S rRNA sequencing, analyzed metagenomes shotgun sequencing. found differences communities' structures...
