Abstract The skeletal ring expansion of heteroarenes through carbene insertion is gaining popularity in synthetic chemistry. Efficient strategies for heterocyclic to access heterocycles containing a fluoroalkyl quaternary carbon center are highly desirable because their broad applications medicinal Herein, we report general strategy the dearomative one‐carbon azoles using N ‐triftosylhydrazones as precursors, resulting ring‐expanded excellent yields with good functional‐group compatibility....

Abstract Given the prevalence of heterocyclic scaffolds in drug‐related molecules, converting these highly modular into structural diversified and dearomatized analogs is an ideal strategy for improving their physicochemical pharmacokinetic properties. Here, we described efficient method silver carbene‐mediated dearomative N−N bond cleavage leading to skeletal hopping between indazole 1,2‐dihydroquinazoline via a selective single‐carbon insertion procedure. Using this methodology, series...

Herein, we developed the new, powerful, and easy-to-handle chemical reagent, dimethoxyacetaldehyde-N-triftosylhydrazone (DMHz-Tfs), as a convenient in situ source of dimethoxydiazoethane under mild conditions. We demonstrate carbene reactivity DMHz-Tfs iron-catalyzed cyclopropanation Doyle-Kirmse reactions, providing access to diverse acetal functionalized cyclopropanes homoallylic- allenyl-sulfides at gram-scale with high stereoselectivity. DFT calculations elucidated involvement most...

Here, we report a silver carbene-enabled single-carbon insertion reaction of indoles via one-pot, two-step sequence to deliver dearomative quaternary center quinoline scaffold in modular fashion. Specifically, used N-triftosylhydrazones as masked donor–donor carbene precursors that facilitate the carbon atoms bearing various functional groups library functionalized quinoline. Experimental and DFT evidence support transient presence cyclopropane species removal protecting groups.

Abstract The cardiotoxicity induced by arsenic trioxide (ATO) limits its clinical application in acute promyelocytic leukemia treatment. Sacubitril/valsartan (LCZ696) is an effective drug for the treatment of heart failure. In this study, we aimed to investigate protective effect and mechanisms LCZ696 against ATO-induced mice H9c2 cells. We found that could alleviate decrease ejection fraction fractional shortening ATO, thereby improving mouse cardiac contractile function. also reduce...
