RASopathies are disorders resulting from mutations in the RAS-MAPK pathway that often cause lymphatic anomalies, including lymphedema, chylous ascites, or chylothorax, which contributes to significant morbidity and mortality. Inhibition of MEK, a crucial enzyme RAS-MAPK-ERK signaling pathway, has been demonstrated modulate upregulated signaling, may lead reconfiguration vessels. While MEK inhibitors have employed off-label for malformations associated with RASopathies, there is limited data...

Cornelia de Lange (CdLS) is caused by genes affecting the function of cohesin that can impact cell replication (mitosis and meiosis) disrupt normal development. Heterozygous pathogenic variants in NIPBL, SMC3, BRD4, RAD21, or hemizygous pathogenetic X-linked HDAC8 SMC1A cause a spectrum severity depending on gene involved and, for some genes, type variant (missense, splice site, frameshift nonsense). RAD21 usually causes milder form CdLS accounts less than 1% all identified variants. Our...
