In the current work we characterize uptake mechanism of two NickFect family members, NF51 and NF1, related to biological activity transfected plasmid DNA (pDNA). Both vectors condense pDNA into small negatively charged nanoparticles that transfect HeLa cells with equally high efficacy delivery is mediated by SCARA3 SCARA5 receptors. NF1 condenses less homogeneous stable than NF51. NF51/pDNA enter via macropinocytosis, while NF1/pDNA complexes use clathrin- or caveolae-mediated endocytosis...
Modifying cell-penetrating peptides (CPPs) with fatty acids has long been used to improve peptide-mediated nucleic acid delivery. In this study we have revisited phenomenon a systematic approach where developed structure–activity relationship describe the role of acyl chain length in transfection process. For that took well-studied CPP, PepFect14, as basis and varied its N-terminal from 2 22 carbons. To evaluate delivery efficiency, were noncovalently complexed splice-correcting...
The CCAAT/enhancer‐binding protein beta (CEBPB or C/EBP β ) is a transcription factor that plays critical role in cellular differentiation, metabolism, and immune response. Emerging evidence has highlighted its complex involvement both solid hematological cancers, such as hepatocellular carcinoma (HCC) pancreatic ductal adenocarcinoma (PDAC), where it can act an oncogene tumor suppressor, depending on the context. In this study, we describe design evaluation of conjugate formed by small...
Biotransformed soybean extract has anti-aromatase and anti-tumoral properties in Estrogen receptor-positive (ER+) breast cancer.
Pancreatic ductal adenocarcinoma (PDAC) is one of the most aggressive malignancies; it preferentially metastasizes to liver and main cause death from this disease. In previous studies, small activating RNA against CCAAT/enhancer-binding protein-α (C/EBPα-saRNA) demonstrated efficacy PDAC in a local subcutaneous tumor model. study, we focused on C/EBPα-saRNA advanced stage PDAC. For targeted delivery, selected new anti-transferrin receptor aptamer (TR14), which high binding affinity target...
Affibody-mediated PNA-based pretargeting is a promising approach to radionuclide therapy of HER2-expressing tumors. In this study, we test the hypothesis that shortening PNA probes would increase tumor-to-kidney dose ratio. The primary probe ZHER2:342-SR-HP15 and complementary secondary HP16, HP17, HP18, containing 9, 12, 15 nucleobases, respectively, carrying 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid (DOTA) chelator were designed, synthesized, characterized in vitro, labeled...
Metabolic syndrome (MetS) is a pathological condition characterized by abdominal obesity, insulin resistance, hypertension, and hyperlipidemia. Sirtuin 1 (SIRT1), highly conserved histone deacetylase, as key metabolic regulator protector against aging-associated pathologies, including MetS. In this study, we investigate the therapeutic potential of activating SIRT1 using small RNAs (saRNA), thereby reducing inflammatory-like responses re-establishing normal lipid metabolism. saRNA...
