Myeloproliferative neoplasms, such as polycythemia vera, essential thrombocythemia, and myelofibrosis, are chronic hematologic cancers with varied progression rates. The genomic characterization of patients myeloproliferative neoplasms offers the potential for personalized diagnosis, risk stratification, treatment.

Clonal proliferation in myeloproliferative neoplasms (MPN) is driven by somatic mutations JAK2, CALR or MPL, but the contribution of inherited factors poorly characterized. Using a three-stage genome-wide association study 3,437 MPN cases and 10,083 controls, we identify two SNPs with significance JAK2(V617F)-negative MPN: rs12339666 (JAK2; meta-analysis P=1.27 × 10(-10)) rs2201862 (MECOM; P=1.96 10(-9)). Two additional SNPs, rs2736100 (TERT) rs9376092 (HBS1L/MYB), achieve when including...

Aims
The Tuberous Sclerosis 2000 Study is the first comprehensive longitudinal study of tuberous sclerosis (TS) and aims to identify factors that determine prognosis. Mode presentation findings at initial assessments are reported here.
Methods
Children aged 0–16 years newly diagnosed with TS in UK were evaluated.
Results
125 children studied. 114 (91%) met clinical criteria for a definite diagnosis remaining 11 (9%) had pathogenic TSC1 or TSC2...

The herpes simplex virus type 1 genome contains four open reading frames (ORFs) which are predicted to encode hydrophobic proteins with the potential cross a membrane several times. products of these genes (genes UL10, UL20, UL43 and UL53) have not previously been identified. To investigate role in life cycle, we attempted inactivate individually by inserting lacZ gene from Escherichia coli within ORFs. Using this approach isolated insertion mutants for UL10 UL43, as well deletion mutant...

Purpose Cytoreductive therapy is beneficial in patients with essential thrombocythemia (ET) at high risk of thrombosis. However, its value those lacking high-risk features remains unknown. This open-label, randomized trial compared hydroxycarbamide plus aspirin alone ET age 40 to 59 years and without factors or extreme thrombocytosis. Patients Methods were lacked a history ischemia, thrombosis, embolism, hemorrhage, thrombocytosis (platelet count ≥ 1,500 × 10 9 /L), hypertension, diabetes...

Background:
Comprehensive diagnostic imaging referral guidelines are an important tool to assist referring clinicians and radiologists in determining the safest best-clinical-value study for their patients; Canadian Association of Radiologists (CAR) last produced its 2012. In partnership with several national organizations, clinicians, radiologists, patient family advisors from across Canada, association is redoing using a new methodology guideline development, these recommendations...

Background Tuberous sclerosis complex (TSC) is associated with intellectual disability, but the risk pathways are poorly understood. Method The Sclerosis 2000 Study a prospective longitudinal study of natural history TSC. One hundred and twenty-five UK children age 0–16 years TSC born between January 2001 December 2006 were studied. Intelligence was assessed using standardized measures at ≥2 age. onset epilepsy, type seizure disorder, frequency duration seizures, as well response to...

We have investigated whether herpes simplex virus (HSV) contains structural polypeptides which are modified by myristic acid. demonstrate that virions contain a family of myristylated proteins, Mr approximately 13,000 to 16,000. These were mapped, using HSV-1/HSV-2 intertypic recombinants, 0.130 0.204 map units on the genome. Using anti-peptide sera, raised against carboxyterminus predicted UL11 gene product, we established virion products viral UL11.

On the basis of predicted amino acid sequence characteristics, herpes simplex virus type 1 gene UL10 is thought likely to encode a membrane protein with eight potential transmembrane regions. Previously, an apparent M(r) 47,000 on SDS-PAGE was identified as product this gene. Here we have further characterized protein, and show that it modified by N-linked glycosylation, associates membranes from infected cells, component particle. It not essential for growth in tissue culture. To...

Herpes simplex virus type 1 (HSV-1) gene UL11 encodes a myristylated virion protein. In this paper we have characterized the product further and investigated its role in life cycle. Wild-type HSV-1 strain 17syn+ expresses three electrophoretically distinguishable polypeptide species. Analysis of single plaque isolates demonstrated that two populations exist within stock: major population encoding only higher Mr species, minor lowest species alone. DNA sequence analysis suggests latter...

We have used antisera raised against synthetic oligopeptides to characterize the protein products from herpes simplex virus type 1 gene US11. These recognized predominantly polypeptides of apparent molecular weight 21,000 and 22,000, but also 17,500, 15,000, 14,000 11,000. Tryptic peptide fingerprint analysis confirmed that these were all closely related. The 22,000 shown be DNA-binding proteins, immune electron microscopy demonstrated their strong localization within nucleoli infected cells.
