Chimeric antigen receptor (CAR) T cells are ineffective against solid tumors with immunosuppressive microenvironments. To overcome suppression, we engineered circuits in which tumor-specific synNotch receptors locally induce production of the cytokine IL-2. These potently enhance CAR cell infiltration and clearance immune-excluded tumors, without systemic toxicity. The most effective IL-2 induction circuit acts an autocrine (TCR)- or CAR-independent manner, bypassing suppression mechanisms...
Resistance to immune checkpoint inhibitors (ICI) is common, even in tumors with T-cell infiltration. We thus investigated consequences of ICI-induced infiltration the microenvironment resistant tumors. T cells and neutrophil numbers increased ICI-resistant following treatment, contrast ICI-responsive Resistant were distinguished by high expression IL1 receptor 1, enabling a synergistic response TNFα induce G-CSF, CXCL1, CXCL2 via NF-κB signaling, supporting immunosuppressive accumulation...

Background: Lymphopenia is associated with poor outcomes and exacerbated by conventional chemoradiation therapies in head neck squamous cell carcinoma (HNSCC). NT-I7 (rhIL-7-hyFc, efineptakin alfa; NeoImmuneTech, Inc.), a long-acting human IL-7, has the potential to restore lymphocyte populations, modulate immune responses, enhance anti-tumor activity. This study investigated NT-I7’s effects on populations its synergy cisplatin checkpoint inhibitors (ICIs) syngeneic, immunocompetent HNSCC...
Using a promiscuous biotin ligase, unique set of force-sensitive proximal proteins surrounding zyxin was identified. Proximal biotinylation, therefore, can be used to detect potential protein interactions.
