Pluripotent stem cells (PSCs) have been successfully developed in many species. However, the establishment of bovine-induced pluripotent (biPSCs) has challenging. Here we report generation biPSCs from bovine mesenchymal (bMSCs) by overexpression lysine-specific demethylase 4A (KDM4A) and other reprogramming factors OCT4, SOX2, KLF4, cMYC, LIN28, NANOG (KdOSKMLN). These exhibited silenced transgene expression at passage 10, had prolonged self-renewal capacity for over 70 passages. The flat,...

Reprogramming of the gamete into a developmentally competent embryo identity is fundamental aspect preimplantation development. One most important processes this reprogramming transcriptional awakening during embryonic genome activation (EGA), which robustly occurs in fertilized embryos but defective somatic cell nuclear transfer (SCNT) embryos. However, little known about genome-wide underlying chromatin landscape EGA SCNT and how it differs from embryo. By profiling open both types bovine...

Bovine in vitro oocyte maturation (IVM) is an easy way to obtain oocytes for subsequent assisted reproductive techniques but inefficient compared vivo maturation. Supplementation of three cytokines, fibroblast growth factor 2 (FGF2), leukemia inhibitory (LIF), and insulin-like 1 (IGF1), or FLI, has increased embryo development multiple species, studies have not explored the differences caused by FLI IVM supplementation. This study aimed assess important nuclear cytoplasmic events...
