We report characterization of hepatitis B virus (HBV) capsids by resistive-pulse sensing through single track-etched conical nanopores formed in poly(ethylene terephthalate) membranes. The pores were ∼40 nm diameter at the tip, and pore surface was covalently modified with triethylene glycol to reduce charge density, minimize adsorption capsids, suppress electroosmotic flow pore. HBV assembled vitro from Cp149, assembly domain capsid protein. Assembled T = 3 (90 Cp149 dimer) 4 (120 are 31 36...

We report fabrication and characterization of nanochannel devices with two nanopores in series for resistive-pulse sensing hepatitis B virus (HBV) capsids. The pores are patterned by electron beam lithography between microchannels etched reactive ion etching. 50-nm wide, deep, 40-nm long spaced 2.0-μm apart. that brackets the is 20× wider (1 μm) to reduce electrical resistance adjacent ensure current returns its baseline value events. Average pulse amplitudes differ <2% demonstrate technique...

ABSTRACT Hepatitis B virus (HBV) capsid proteins (Cps) assemble around the pregenomic RNA (pgRNA) and viral reverse transcriptase (P). pgRNA is then transcribed to double-stranded DNA (dsDNA) within capsid. The Cp assembly domain, which forms shell of capsid, regulates kinetics stability. Cp, via its nucleic acid-binding C-terminal also affects acid organization. We hypothesize that structure may have a direct effect on processing. Using structure-guided design, we made series mutations at...

ABSTRACT Capsid (core) assembly is essential for hepatitis B virus (HBV) replication. We hypothesize that kinetics and stability are tuned optimal viral replication, not maximal assembly. Assembly effectors (AEfs) small molecules proposed to disrupt this balance by inappropriately enhancing core Guided the structure of an AEf-bound core, we designed a structural mimic protein, V124W mutant. In biochemical studies, mutant recapitulated effects AEfs, with fast strong protein-protein...

A Conversational Recommender System (CRS) offers increased transparency and control to users by enabling them engage with the system through a real-time multi-turn dialogue. Recently, Large Language Models (LLMs) have exhibited an unprecedented ability converse naturally incorporate world knowledge common-sense reasoning into language understanding, unlocking potential of this paradigm. However, effectively leveraging LLMs within CRS introduces new technical challenges, including properly...

Woodchuck hepatitis virus (WHV), a close relative of human B (HBV), has been key model for disease progression and clinical studies. Sequences the assembly domain WHV HBV core proteins (wCp149 hCp149, respectively) have 65% identity, suggesting similar behaviors. We report cryo-electron microscopy (cryo-EM) structure capsid at nanometer resolution characterization wCp149 assembly. At this resolution, T=4 structures are practically identical. In contrast to their structural similarity,...

Abstract: The alphavirus Chikungunya virus is transmitted to humans via infected mosquitos. Most experience symptoms which can range from short-term fatigue and fever debilitating arthritis that last for months or years. Some patients relapse years after the initial bout of disease. capsid protein forms a shell around viral RNA genome; this structure called nucleocapsid core. core protects genome during transmission with correct environmental trigger, proteinaceous dissociates releases...

Wideband oscillations seriously threaten the stable operation of power systems. The wind farm modeling is basis wideband oscillation analysis, but accurate model high order and not available, impeding analysis oscillations. Hence, this paper proposes an equivalent method based on speed-frequency two-variable admittance model. Firstly, relationships between steady-state variables permanent magnetic synchronous generator turbine (PMSG-WT) speed are deduced. Then, can be obtained. Finally, in...
